技术领域
本发明涉及海棠果生物多酚及其制备方法。
背景技术
海棠果多酚主要由具有生物活性的儿茶素、表儿茶素、原花青素、根皮苷、根皮素、槲皮素等黄酮组成。花青素等黄烷-3醇结构的低聚及高聚黄酮类化合物，作为一种天然存在的强效抗氧化剂，具有显著清除人体内自由基功能，医学研究证明人类有70多种疾病与自由基有关，如：心血管类、癌症、类风湿、早衰等，现代药理学研究发现，具有黄烷-3醇结构的多酚还具有显著的降低低密度脂蛋白的作用。槲皮素作为海棠果中的主要活性物质，药理学研究表明具有抗肿瘤、抗氧化、抗血栓、抗糖尿病及保护心血管等功效。根皮素和根皮苷具有平缓降血压和降糖等作用，另根皮苷具有美白作用。然而，现有技术获得的海棠果多酚水溶性差、多酚含量低，不能作为功能性饮料、化妆品等水基性产品的添加剂。
发明内容
本发明所要解决的技术问题是提供一种水溶性得到改善的海棠果多酚的制备方法及其应用。
本发明以野生或家种的海棠果为原料，应用膜分离技术去除提取液中的生物大分子物质，特别是功能活性不高的大分子聚合鞣质，使海棠果提取液在水中的溶解性得到很大改善。经使用HUNTER LAB仪器检测，由提取物配成的0.1%水溶液的L值可以达到93以上。若进一步将海棠果提取液经大孔树脂吸附纯化，则制备得到的海棠果多酚提取物的生物多酚含量得到显著提高，可达到85%以上。
本发明解决其技术问题所采取的技术方案是：该海棠果多酚的制备方法主要包括以下步骤：
1)以海棠果为原料提取得到海棠果的提取液；
2)用纳米滤膜分离海棠果的提取液以去除缩合鞣质和生物大分子物质，得到过滤后的海棠果的提取液；所述纳米滤膜为能够截留分子量大于2000Da的物质的高分子材料纳米滤膜或无机陶瓷纳米滤膜；
3）将得到的海棠果的提取液进行减压浓缩，将浓缩液喷雾干燥得到海棠果多酚提取物成品。
进一步地，本发明在所述步骤2）和步骤3）之间还包括以下步骤：
将步骤2）中所得到的过滤后的海棠果的提取液先用大孔树脂进行吸附，然后水洗脱以去除杂质，再使用低级醇水溶液进行解吸附，得到解吸附后的海棠果的提取液。
进一步地，在本发明所述低级醇水溶液中，低级醇与水的体积比为70~95%。
进一步地，本发明所述低级醇为甲醇、乙醇、异丙醇或正丁醇。
进一步地，本发明所述大孔树脂为聚苯丙烯、或为聚苯乙烯与功能基团交联的树脂。
本发明的海棠果多酚作为添加剂的应用是：所述海棠果多酚作为水基性产品的添加剂。
进一步地，本发明所述水基性产品为功能性饮料或化妆品。
本发明的优点是：（1）制备得到的海棠果多酚提取物可以用于食品抗氧化剂，其抗氧化能力是工业抗氧化剂（BHT）的20倍，可以用于代替BHT添加到食品、油类等产品中；可作为保健食品原料开发出具有清除自由基、美容养颜的保健食品；可进一步开发得到具有降血压和将血糖的中药五类新药（中药五类新药一般要求一类物质含量达到50%以上，而按照本发明的方法，总多酚能够达到85%以上）。（2）本发明方法制备的海棠果多酚水溶性好、多酚含量高；可作为功能饮料和水基化妆品的活性添加剂，从而生产出总多酚含量达到85%以上的饮料专用型海棠多酚提取物产品，产品的水溶性用HUNTER LAB仪器检测，L值高于93以上。（3）本发明方法简单，易于操作，工业化生产具有可操作性，得到的海棠果多酚产品，用福林酚试剂方法评价，其含量可以达到65%以上，并且能够通过一步精制获得含量达到85%的海棠果多酚提取物。
具体实施方式
作为本发明的优选实施方式，海棠果总多酚的制备方法包括以下步骤：
（1）将海棠果原料用压榨机压榨，或用水提取，过滤得到海棠果汁；
（2）用能够截留分子量大于2000Da的物质的高分子材料纳米滤膜或陶瓷纳米滤膜进行分离去除缩合鞣质和生物大分子物质，得到过滤后的海棠果的提取液；
（3）将步骤（2）得到的纳滤液，再经大孔树脂进行吸附，用低级醇水溶液进行解吸附，精制得到高多酚含量的海棠果多酚提取物。其中，大孔树脂优选聚苯乙烯骨架或聚苯丙稀骨架材料大孔树脂，其平均孔径约为
比表面为400-760m2/g。例如可选用罗门哈斯的XAD-7，或者沧州宝恩的HPD100C型树脂。
纳米滤膜为能够截留分子量大于2000Da的物质的高分子材料纳米滤膜或无机陶瓷纳米滤膜。例如可选用美国海德能公司（HYDRANAUTICS）的截留分子量2000Da以上的纳滤膜产品。此外，安徽世杰公司的纳滤陶瓷膜也可以达到分离海棠果多酚的目的。
由于海棠果多酚在水以及低级醇中溶解性好，而其脂溶性非常差，因此使用高级醇洗脱效果不好，因此，为达到较好的解吸附效果，本发明使用低级醇水溶液进行洗脱。在低级醇水溶液中，低级醇可为甲醇、乙醇、异丙醇、正丁醇或其它醇，其中，低级醇与水的体积比为70~95%时具有最佳的洗脱效果。
（4）将步骤（3）解吸附得到的溶液进行减压浓缩，将浓缩液喷雾干燥得到海棠果多酚提取物成品。
作为本发明的具体实施方式，可在海棠果生长的各个生长阶段（果实含量5克、10克、20克、50克）进行采摘，破碎，用2－5倍体积比热水渗滤，得到海棠果提取汁，浓缩得到浓缩汁(糖度30—65)；醇沉，过滤去除沉淀，将上清液用纳滤膜分离，截留分子量在2000Da以上的物质（例如缩合鞣质和生物大分子物质等），浓缩透过纳滤膜的乙醇溶液，喷雾干燥得到带有海棠特有酸涩味的提取物，其中多酚含量在50—70%之间。
将透过纳滤膜的乙醇溶液，浓缩去除乙醇，上聚丙烯结构的大孔吸附树脂，优选罗门哈斯XAD-7的大孔树脂，水冲洗去除海棠果中的苹果酸，氨基酸等物质，用乙醇洗脱，浓缩，喷雾干燥，得到海棠特有涩味的提取物，使多酚含量提高到70-85%，其水溶性用HUNTER LAB仪器检测，0.1%水溶液的L值高于93以上。
如果所需的海棠果多酚提取物成品的多酚含量为65-85%，则可以不必对纳米滤膜过滤后的海棠果提取汁进行吸附和解吸附，而直接进行浓缩和喷雾干燥即可。
以下以具体的实施例进一步说明本发明的海棠果多酚的制备方法。

取棠果（平均重量在15克左右/个，河北怀来7月10日采摘）1000公斤，用袋式压榨机，得到海棠果汁600升（相对密度为1.01，60度），过120目滤布后，用板框过滤器过滤，得到澄清液体590升，浓缩得到相对密度1.08浓缩液40升；加入95%体积比乙醇200升，搅拌，静置冷却12小时，吸取上清液体通过可截留分子量在2000Da以上的物质的纳米滤膜，继续用200升水冲洗膜，得到380升液体；浓缩去除乙醇，以每小时150升的流速通过体积为200升的大孔树脂进行吸附。吸附完成后，用500升体积的去离子水进行冲洗，去除杂质；用70%乙醇的水溶液200升进行解吸附，流速100升/小时，最终得到250升体积的洗脱液体，在负压0.75大气压、温度低于65摄氏度的条件下减压浓缩得到相对密度为1.08的海棠果浓缩汁，体积为51升；将浓缩汁喷雾干燥得到成品11.3公斤。用福林酚法检测多酚的含量为88.9%；0.1%水溶液的L值为93.5。
实施例2：
取海棠果（平均重量在20克左右/个，河北怀来8月3日采摘）500公斤，用袋式压榨机，得到海棠果汁300升（相对密度为1.01），过120目滤布后，用板框过滤器过滤，得到澄清液体280升，浓缩得到相对密度为1.08的浓缩液21升；再加入95%体积比乙醇100升，搅拌，静置冷却12小时，吸取上清液体通过可截留分子量在2000Da以上的物质的纳米滤膜，继续用100升水冲洗膜，得到180升液体；浓缩去除乙醇，以每小时70升的流速通过体积为100升的大孔树脂，进行吸附。吸附完成后，用200升体积的去离子水进行冲洗，去除杂质；用100升的95%乙醇进行解吸附，流速100升/小时，最终得到150升体积的洗脱液体，在负压0.75大气压、温度低于65摄氏度的条件下减压浓缩得到相对密度为1.07的海棠果浓缩汁，体积为24升；将浓缩汁喷雾干燥得到成品4.3公斤。用福林酚法检测多酚的含量为87.9%；0.1%水溶液的L值为94.2。
实施例3：
取海棠果（平均重量在5克左右/个,河北怀来5月20日采摘）100公斤，用500公斤80度的水提取2小时，过滤得到海棠汁450升（相对密度1.01），过120目滤布后，用板框过滤器过滤，得到澄清液体440升，提取液通过纳滤膜，继续用100升水冲洗膜，得到580升液体；在负压0.75大气压、温度低于65摄氏度的条件下减压浓缩得到相对密度为1.08的海棠果浓缩汁，体积为15升；将浓缩汁喷雾干燥得到成品3公斤，用福林酚法检测多酚含量为68.9%。0.1%水溶液的L为93.2。
由本发明制备得到的海棠果多酚提取物可以用于食品抗氧化剂，其抗氧化能力是工业抗氧化剂（BHT）的20倍，可以用于代替BHT添加到食品、油类等产品中；可作为保健食品原料开发出具有清除自由基、美容养颜的保健食品；可进一步开发得到具有降血压和降血糖的中药五类新药（中药五类新药一般要求一类物质含量达到50%以上，而按照本发明的方法，总多酚能够达到85%以上）。本发明方法制备的海棠果多酚水溶性好、多酚含量高；可作为功能饮料和水基化妆品的活性添加剂，从而生产出总多酚含量达到85%以上的饮料专用型海棠多酚提取物产品，产品的水溶性用HUNTER LAB仪器检测，L值高于93以上。本发明方法简单，易于操作，工业化生产具有可操作性，得到的海棠果多酚产品，用福林酚试剂方法评价，其含量可以达到65%以上，并且能够通过一步精制获得含量达到85%的海棠果多酚提取物。
Claims (6)
1.一种海棠果多酚的制备方法，其特征是包括以下步骤：
1) 以海棠果为原料提取得到海棠果的提取液；
2) 用纳米滤膜分离海棠果的提取液以去除缩合鞣质和生物大分子物质，得到过滤后的海棠果的提取液；所述纳米滤膜为能够截留分子量大于2000Da的物质的高分子材料纳米滤膜或无机陶瓷纳米滤膜；
3）将得到的海棠果的提取液进行减压浓缩，将浓缩液喷雾干燥得到海棠果多酚提取物成品。
2.根据权利要求1所述的海棠果多酚的制备方法，其特征是在所述步骤2）和步骤3）之间还包括以下步骤：
将步骤2）中所得到的过滤后的海棠果的提取液先用大孔树脂进行吸附，然后水洗脱以去除杂质，再使用低级醇水溶液进行解吸附，得到解吸附后的海棠果的提取液；所述低级醇为甲醇、乙醇、异丙醇或正丁醇。
3.根据权利要求2所述的海棠果多酚的制备方法，其特征是：在所述低级醇水溶液中，低级醇与水的体积比为70~95%。
4.根据权利要求2所述的海棠果多酚的制备方法，其特征是：所述大孔树脂为聚苯丙烯、或为聚苯乙烯与功能基团交联的树脂。
5.一种将权利要求1的海棠果多酚作为添加剂的应用，其特征是：所述海棠果多酚作为水基性产品的活性添加剂。
6.根据权利要求5所述的海棠果多酚作为添加剂的应用，其特征是：所述水基性产品为功能性饮料或化妆品。
