Abstract
本发明涉及一种尿苷的生产方法,利用分子生物学方法向Bacillus subtilis A219的氨甲酰磷酸合成酶大亚基中引入突变位点(949位谷氨酸缺失,E949*)获得工程菌Bacillus subtilis A2191;在发酵罐中对工程菌进行发酵培养获得尿苷发酵液,工程菌的尿苷产量可以达到40g/L以上;从尿苷发酵液中分离提取得到尿苷晶体,晶体纯度大于98%。本发明利用微生物发酵法实现了尿苷的生产,具有生产成本低、污染小、产品收率和纯度高的优势,具备较高的工业推广应用价值。

技术领域
本发明涉及生物制药技术领域,尤其是一种尿苷的生产方法。
背景技术
尿苷是一种重要的嘧啶核苷,可作为前体用于制备核苷类药物,如抗肿瘤药物5-氟尿苷,5-氟尿嘧啶核苷,抗病毒药物5-碘脱氧尿苷等。尿苷还可以用于制备尿苷酸钠,后者可以作为调味品添加到牛奶中以增强婴儿的免疫力,或者用于合成肝脏解毒物质葡萄糖醛酸。
传统的尿苷生产方法是使用核糖核酸(RNA)经过化学方法水解,然后分离提纯得到尿苷和胞苷,这不仅涉及大量的优质RNA原料,而且两种嘌呤核苷和两种嘧啶核苷必须产销平衡,否则成本价会很高。与化学法相比,用选育获得的特殊性状的菌株一步发酵法生产尿苷具有生产成本低,环境污染小,分离提纯简单等优势。日本武田化学公司在20世纪80年代开展了一系列的发酵法生产尿苷的研究工作,通过化学诱变获得了具有尿嘧啶缺陷、6-氮杂尿嘧啶抗性以及尿苷磷酸化酶活性丧失的菌株No.556,能够积累55g/L尿苷(Appliedmicrobiology and biotechnology,1989,30(3):234-238.)。进一步对培养条件以及培养基成分优化,在6000L的发酵罐中该菌株能稳定生产65g/L尿苷(Bioscience,biotechnology,and biochemistry,1994,58(9):1608-1612.)。而国内对于尿苷的发酵法生产仍停留在研发阶段。
前期的发明中,申请人通过常压等离子诱变结合嘧啶核苷结构类似物筛选获得高产嘧啶核苷的诱变菌株A219(CN 105671008 A)和A260(CN 105671007 A),并发现了氨甲酰磷酸合成酶的变构调节位点,但诱变菌株的尿苷产量仍需进一步提高。
发明内容
本发明所要解决的技术问题在于提供一种尿苷的生产方法。
为解决上述技术问题,本发明的技术方案是:
一种尿苷的生产方法,利用分子生物学方法向Bacillus subtilis A219的氨甲酰磷酸合成酶大亚基中引入突变位点(949位谷氨酸缺失,E949*)获得工程菌Bacillussubtilis A2191;在发酵罐(5L)中对工程菌进行发酵培养获得尿苷发酵液;从尿苷发酵液中分离提取得到尿苷晶体;所述Bacillus subtilis A219保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏编号为CGMCC No.11774,该菌株氨甲酰磷酸合成酶大亚基具有序列表1所述核苷酸序列、序列表2所述氨基酸序列,其第1016位氨基酸为亮氨酸。
优选的,上述尿苷的生产方法,所述分子生物学方法是以Bacillus subtilisA219氨甲酰磷酸合成酶大亚基基因为模板,设计合成包含E949*突变位点的基因片段(见序列表8所示序列),再通过原位修饰的方法使用合成的基因片段替换Bacillus subtilisA219的氨甲酰磷酸合成酶大亚基基因中的同源基因片段,从而获得工程菌Bacillussubtilis A2191。工程菌Bacillus subtilis A2191中氨甲酰磷酸合成酶大亚基应包含E949*突变位点(核苷酸序列见序列表3,氨基酸序列见序列表4)。
优选的,上述尿苷的生产方法,所述发酵培养方法是将工程菌Bacillus subtilisA2191接入到发酵培养基中,接种量为10%-15%,培养温度为34-38℃,pH值为6.7-7.0,相对溶氧控制在10%-30%,发酵过程中流加800g/L葡萄糖溶液并控制残糖浓度为0.5%-2%,发酵周期为45-65h。发酵完成后,工程菌A2191尿苷产量可以达到40g/L以上。
优选的,上述尿苷的生产方法,所述pH值是通过流加25%的氨水和玉米浆进行控制。
优选的,上述尿苷的生产方法,所述相对溶氧值是通过调节通气量和搅拌转速进行控制。
优选的,上述尿苷的生产方法,所述发酵培养基组成为:葡萄糖50g/L,酵母粉5g/L,KH2PO4 1g/L,K2HPO4 5.2g/L,(NH4)2SO4 5g/L,MgSO4·7H2O 5g/L,MnSO4 0.02g/L,ZnSO40.02g/L,柠檬酸钠10g/L,谷氨酸钠10g/L。
优选的,上述尿苷的生产方法,初始葡萄糖,流加葡萄糖(800g/L)以及流加玉米浆单独灭菌,其余组分一起灭菌,灭菌条件为115℃,15min。
优选的,上述尿苷的生产方法,所述尿苷分离提取方法(从尿苷发酵液中分离提取得到尿苷晶体)是指收集工程菌发酵液,离心方法去除菌体后,使用超滤膜过滤方法除去大分子杂质,再使用串联离子交换树脂方法对膜过滤液进行脱色除杂,最后收集树脂流出液进行浓缩结晶获得尿苷成品。
优选的,上述尿苷的生产方法,所述离心方法是使用高速离心机,离心转速8000-10000r/min,离心时间10-15min。
优选的,上述尿苷的生产方法,所述超滤膜过滤方法是使用截留分子量500-1000Da的有机中空纤维超滤膜,操作压力0.5-1.5Mpa,操作温度30-40℃。
优选的,上述尿苷的生产方法,所述串联离子交换树脂方法是将超滤膜过滤液依次通过弱碱性大孔型阴离子交换树脂和强酸性大孔型阳离子交换树脂,两种树脂的装填体积相同,进料浓度(尿苷浓度)为35-40g/L,进料体积为1-2BV,进料流速为0.5-1BV/h。
优选的,上述尿苷的生产方法,所述浓缩结晶是使用真空浓缩将树脂流出液浓缩至过饱和状态,再放入结晶器中进行降温结晶,用无水乙醇洗晶2-3次,得到的尿苷晶体的纯度大于98%。
本发明的有益效果是:
1、本发明在现有技术的基础上,通过向Bacillus subtilis A219(CGMCCNo.11774)氨甲酰磷酸合成酶大亚基中引入突变位点(949位谷氨酸缺失),进一步解除了从头合成途径中尿苷酸对于氨甲酰磷酸合成酶的反馈抑制,显著提升了尿苷的发酵产量。
2、本发明设计了适用于尿苷的分离提取方法,由于尿苷分子呈弱电性,不能被弱碱性大孔型阴离子交换树脂以及强酸性大孔型阳离子交换树脂吸附,因此可以用上述两种树脂串联,对发酵液中的色素、残留的无机盐以及小分子杂质进行吸附,进而高效快速的获得高纯度的尿苷成品。
综上,上述尿苷的生产方法,为了进一步提高诱变菌株的尿苷产量,克服现有技术尿苷产量低,难以分离的缺陷,利用分子生物学技术向A219的氨甲酰磷酸合成酶中引入新的突变位点获得新的工程菌,并提供了工程菌的发酵和尿苷的分离提取方法,为工业化尿苷的生产提供了参考依据。

为了使本领域的技术人员更好的理解本发明的技术方案,下面结合具体实施方式对本发明所述技术方案作进一步的详细说明。其中,

实施例1.工程菌Bacillus subtilis A2191的构建
(1)以Bacillus subtilis A219氨甲酰磷酸合成酶大亚基基因为模板(核苷酸序列见序列表1,氨基酸序列见序列表2),设计并委托金唯智公司合成包含E949*突变位点的基因片段M(见序列表8)。
(2)通过重叠PCR方法,拼接一段由上游同源臂U(见序列表5)、氯霉素抗性基因cat(见序列表7)和下游同源臂D(见序列表6)构成的DNA片段,转化受体菌Bacillus subtilisA219感受态细胞,通过同源重组,在氨甲酰磷酸合成酶大亚基基因(核苷酸序列见序列表1,氨基酸序列见序列表2)位点第2846碱基附近,插入氯霉素抗性基因(基因序列见序列表7),并替换掉部分基因编码序列。
(3)通过筛选氯霉素抗性菌株,获得尿嘧啶缺陷型转化子,将(1)中扩增得到的基因片段M(见序列表8)与上游同源臂U’(见序列表5)和下游同源臂D’(见序列表6)拼接成U’MD’片段,转化尿嘧啶缺陷株,通过同源重组,M片段将替换掉氯霉素抗性基因cat。用基本培养基平板筛选转化子,得到原养型的工程菌Bacillus subtilis A2191。经过测序验证工程菌Bacillus subtilis A2191中氨甲酰磷酸合成酶大亚基应包含E949*突变位点(基因序列见序列表3,氨基酸序列见序列表4)。
(4)上述基因扩增所用引物的序列表如表1所示。

实施例2.利用工程菌Bacillus subtilis A2191发酵生产尿苷
(1)活化培养:取保藏的工程菌Bacillus subtilis A2191划线接种于活化斜面,37℃培养12h,并传代一次。用接种环刮取一环斜面种子接种于装有100mL种子培养基的1L三角瓶中,九层纱布封口,37℃,200rpm振荡培养6h。
(2)发酵培养:将活化的种子接入到含有3L发酵培养基的5L发酵罐中,接种量为10%,培养温度为36℃,相对溶氧控制在20%,发酵过程中流加800g/L葡萄糖溶液并控制残糖浓度为1%,,流加25%的氨水和玉米浆控制发酵过程pH值为7.0,发酵周期为60h。
(3)产物检测:发酵结束后,使用高效液相色谱仪测定发酵液中尿苷浓度。所用色谱柱为Phenomenex Gemini 5u C18 110A 150*4.6mm,柱温30℃,流动相为乙腈:水=2:98,流速1mL/min,每个样品进样10μL,分析时间15min。经测定发酵液中尿苷浓度可达42g/L。
斜面培养基:葡萄糖1g/L,蛋白胨10g/L,牛肉膏g/L,酵母粉5g/L,NaCl 2.5g/L,琼脂20g/L,pH 6.7-7.0。灭菌条件为115℃,15min.
种子培养基:葡萄糖40g/L,酵母粉5g/L,蛋白胨10g/L,NaNO3 15g/L,KH2PO4 1g/L,K2HPO4 5.2g/L,MgSO4·7H2O 1g/L,柠檬酸钠1g/L,pH 6.7~7.0。灭菌条件为115℃,15min。
发酵培养基:葡萄糖50g/L,酵母粉5g/L,KH2PO4 1g/L,K2HPO4 5.2g/L,(NH4)2SO45g/L,MgSO4·7H2O 5g/L,MnSO4 0.02g/L,ZnSO4 0.02g/L,柠檬酸钠10g/L,谷氨酸钠10g/L。其中,初始葡萄糖,流加葡萄糖(800g/L)以及流加玉米浆单独灭菌,其余组分一起灭菌。灭菌条件为115℃,15min。
实施例3.从发酵液中分离提取尿苷
(1)收集发酵液,高速离心(8000r/min,15min)去除菌体。
(2)使用截留分子量600Da的有机中空纤维超滤膜处理离心上清液,除去上清液中的大分子可溶性蛋白和色素,操作压力为1.0Mpa,操作温度40℃。
(3)将超滤膜过滤液依次通过弱碱性大孔型阴离子交换树脂D301和强酸性大孔型阳离子交换树脂D001,除去色素、残留的无机盐以及小分子杂质。两种树脂的装填体积相同,进料浓度(尿苷浓度)为38g/L,进料体积为1.5BV,进料流速为0.75BV/h。
(4)将树脂流出液真空浓缩至过饱和状态,再放入结晶器中进行降温结晶获得尿苷晶体,用无水乙醇洗晶2次。经检测,尿苷晶体的纯度为98.5%。
上述参照具体实施方式对该一种尿苷的生产方法进行的详细描述,是说明性的而不是限定性的,可按照所限定范围列举出若干个实施例,因此在不脱离本发明总体构思下的变化和修改,应属本发明的保护范围之内。

Claims (10)
1.一种尿苷的生产方法,其特征在于:利用分子生物学方法向Bacillus subtilisA219的氨甲酰磷酸合成酶大亚基中引入突变位点获得工程菌Bacillus subtilis A2191;在发酵罐中对工程菌进行发酵培养获得尿苷发酵液;从尿苷发酵液中分离提取得到尿苷晶体;所述Bacillus subtilis A219保藏于中国微生物菌种保藏管理委员会普通微生物中心,保藏编号为CGMCC No.11774,该菌株氨甲酰磷酸合成酶大亚基具有序列表1所述核苷酸序列、序列表2所述氨基酸序列,其第1016位氨基酸为亮氨酸。
2.根据权利要求1所述的尿苷的生产方法,其特征在于:所述分子生物学方法是以Bacillus subtilis A219氨甲酰磷酸合成酶大亚基基因为模板,设计合成包含E949*突变位点的基因片段、见序列表8所示序列,再通过原位修饰的方法使用合成的基因片段替换Bacillus subtilis A219的氨甲酰磷酸合成酶大亚基基因中的同源基因片段,从而获得工程菌Bacillus subtilis A2191;工程菌Bacillus subtilis A2191中氨甲酰磷酸合成酶大亚基应包含E949*突变位点,其核苷酸序列见序列表3,氨基酸序列见序列表4。
3.根据权利要求1所述的尿苷的生产方法,其特征在于:所述发酵培养方法是将工程菌Bacillus subtilis A2191接入到发酵培养基中,接种量为10%-15%,培养温度为34-38℃,pH值为6.7-7.0,相对溶氧控制在10%-30%,发酵过程中流加800g/L葡萄糖溶液并控制残糖浓度为0.5%-2%,发酵周期为45-65h。
4.根据权利要求3所述的尿苷的生产方法,其特征在于:所述pH值是通过流加25%的氨水和玉米浆进行控制。
5.根据权利要求3所述的尿苷的生产方法,其特征在于:所述相对溶氧值是通过调节通气量和搅拌转速进行控制。
6.根据权利要求3所述的尿苷的生产方法,其特征在于:所述发酵培养基组成为:葡萄糖50g/L,酵母粉5g/L,KH2PO4 1g/L,K2HPO4 5.2g/L,(NH4)2SO4 5g/L,MgSO4·7H2O 5g/L,MnSO4 0.02g/L,ZnSO4 0.02g/L,柠檬酸钠10g/L,谷氨酸钠10g/L。
7.根据权利要求3所述的尿苷的生产方法,其特征在于:所述尿苷分离提取方法是收集工程菌发酵液,离心方法去除菌体后,使用超滤膜过滤方法除去大分子杂质,再使用串联离子交换树脂方法对膜过滤液进行脱色除杂,最后收集树脂流出液进行浓缩结晶获得尿苷成品。
8.根据权利要求3所述的尿苷的生产方法,其特征在于:所述离心方法是使用高速离心机,离心转速8000-10000r/min,离心时间10-15min;所述超滤膜过滤方法是使用截留分子量500-1000Da的有机中空纤维超滤膜,操作压力0.5-1.5Mpa,操作温度30-40℃。
9.根据权利要求7所述的尿苷的生产方法,其特征在于:所述串联离子交换树脂方法是将超滤膜过滤液依次通过弱碱性大孔型阴离子交换树脂和强酸性大孔型阳离子交换树脂,两种树脂的装填体积相同,进料浓度为35-40g/L,进料体积为1-2BV,进料流速为0.5-1BV/h。
10.根据权利要求7所述的尿苷的生产方法,其特征在于:所述浓缩结晶是使用真空浓缩将树脂流出液浓缩至过饱和状态,再放入结晶器中进行降温结晶,用无水乙醇洗晶2-3次。
