Abstract
本发明涉及一种降血脂的中药组合物及其制法,该中药组合物包括如下重量配比的原料组成:丹参140~160份、姜黄140~160份、枳壳140~160份、鸡血藤140~160份、黄芪90~110份、毛冬青90~110份、天麻80~100份、大黄80~100份、丝瓜络80~100份、桂枝80~100份、僵蚕50~70份、全蝎50~70份;该中药组合物具有行气止痛,化瘀通络,补气养血,化痰开窍之功,临床上对于高血脂的患者具有显著的疗效。

技术领域
本发明涉及一种降血脂的中药组合物及其制法,属于中药技术领域。
背景技术
随着我国经济的持续发展,人民群众的生活水平逐渐提高,饮食和生活方式也开始向多元化方向发展,近年来,我国人口血脂水平呈现出逐年上升的趋势,高血脂患者也随之大幅增加。尤其是在老年人群中,高血脂更是成为了常见病和多发病,严重威胁着人们的身体健康。
高血脂症是一种常见而多发的代谢性疾病,是引起心脑血管疾病的重要危险因素。中医学上无高血脂症的病名,认为高血脂症为本虚标实,属“血瘀”、“痰浊”范畴,临床辨证多归于肝肾亏虚、脾虚痰浊、气滞血瘀,治疗时大多采用补益肝肾、健脾益气、滋阴养血、活血化瘀、清热通便、消食化痰之药。
目前对于高血脂症的患者常用口服药物有洛伐他汀,阿托伐他汀等,但这些药物长期服用会产生一定的耐药性,导致其治疗效果不佳。若长期服用此类药物,还会才产生较大的的毒副作用,损害肝肾等脏器。然而,中医药对此类疾病的治疗和预防有着巨大的优势。
发明内容
本发明目的在于提供一种降血脂的中药组合物及其制法,该中药组合物对于临床高血脂患者,具有疗效显著,毒副作用小,生产工艺技术简单可行,生产成本低,患者服用方便的优点。
本发明所述降血脂的中药组合物,它包括如下重量配比的原料组成:丹参140~160份、姜黄140~160份、枳壳140~160份、鸡血藤140~160份、黄芪90~110份、毛冬青90~110份、天麻80~100份、大黄80~100份、丝瓜络80~100份、桂枝80~100份、僵蚕50~70份、全蝎50~70份。
该中药组合物的具体原料组方配比为:丹参150份、姜黄150份、枳壳150份、鸡血藤150份、黄芪100份、毛冬青100份、天麻90份、大黄90份、丝瓜络90份、桂枝90份、僵蚕60份、全蝎60份。

本发明中药组合物的制备方法如下:取上述药材,采用真空微波干燥法进行干燥,干燥温度为60~70℃,干燥时间为0.5-2小时,并采用超微粉碎成100~300目细粉,混合均匀后,再加入药剂学上常用的辅料制成各种不同口服制剂,即得。
本方在临床上长期应用治疗高血脂症,取得良好的疗效。方中丹参,性苦,微寒,善能通行血脉,祛淤止痛,且祛淤生新而不伤正,为调理血分之首药。姜黄,可破血行气,通经止痛,与丹参相配合用,加强方药行气止痛之功;配以枳壳可疏肝行气。以上三药共为君药。鸡血藤,苦温而不燥,温而不烈,长于行血散瘀,舒筋活络,又兼补血之功,配以黄芪,力专性走,大补元气,使气旺则血行。又辅以毛冬青,大黄,辅助君药活血通脉;加以天麻熄风止痉,可治头风,四肢强痉,语言不顺;丝瓜络,甘,平。通络,活血,祛风。加以桂枝温通,入营血,通血脉,可使诸药上行于心,脑。配合僵蚕,全蝎,疏通经络而止痛。上述诸药共为臣药;再辅以伸筋草,络石藤,能通行经络,熄风止痉。当归可补血活血,补中有动,为血中之圣药。石菖蒲,辛温,可开窍豁痰,交通心肾,痰化窍开;泽泻,渗湿泻热,导热从水道而去。最后,配以桑寄生,可补益肝肾,以治本。上述诸药共为佐使药,全方诸药合用,可使得血脉相通,气机调畅,诸症自愈。全方共奏行气止痛,化瘀通络,补气养血,化痰开窍之功。本方配伍的特点,处方活血中配以行气,即能散瘀止痛,又可解气滞;祛瘀与补血兼顾,可达标本兼治之功。并起到治疗高血症的目的。

为了说明本发明降血脂技术方案的整体有益效果,我们进行了一系列的药学对比研究,具体内容为:
试验药物1组:取丹参150g、姜黄150g、枳壳150g、鸡血藤150g、黄芪100g、毛冬青100g、天麻90g、大黄90g、丝瓜络90g、桂枝90g、僵蚕60g、全蝎60g,采用真空微波干燥法进行干燥,干燥温度为65℃,干燥时间为1小时,并采用普通粉碎成100目细粉,混合均匀后,再加入药剂学上常用的淀粉制成胶囊剂,即得。
试验药物2组:取丹参150g、姜黄150g、枳壳150g、鸡血藤150g、黄芪100g、毛冬青100g、天麻90g、大黄90g、丝瓜络90g、桂枝90g、僵蚕60g、全蝎60g、伸筋草60g、当归60g、络石藤60g、刺五加60g、石菖蒲60g、泽泻60g、桑寄生60g,采用真空微波干燥法进行干燥,干燥温度为65℃,干燥时间为1小时,并采用普通粉碎成100目细粉,混合均匀后,再加入药剂学上常用的淀粉制成胶囊剂,即得。
本发明中药有效成分测定方法如下:
⑴、水溶性浸出物测定法:取本发明实验药物1组和2组的中药混合粉末约3g,称定重量置250ml的锥形瓶中,精密加入水100ml,摇匀塞紧,称定重量,静置1小时后,连接回流冷凝管,加热至沸腾,并保持微沸1小时,放冷后,取下锥形瓶,密塞,称定重量,用水补足减失的重量,摇匀,用干燥滤器滤过,精密量取滤液25ml,置已干燥至恒重的蒸发皿中,在水浴上蒸干后,于105℃干燥3小时后,移置干燥器中,冷却30分钟,迅速精密称定重量,除另有规定外,以干燥品计算本发明中药组合物中水溶性浸出物的百分数。
⑵、醇溶性浸出物测定法:照上述测定方法⑴水溶性浸出物测定法测定,以各该品种项下规定50%乙醇代替水为溶剂。

⑶、本发明中药组合物中有效成分含量测定
采用高效液相色法,用Agilent ZORBAX Ec lipse XDB-C18(4.6mm×150mm,5μm),以甲醇-水(15∶5)为流动相,流速1mL/min,检测波长270nm,对本发明中药组合物中丹参酮ⅡA的含量进行测定;以紫外分光光度法测定波长采用香草醛-高氯酸作为显色体系,紫外分光光度法测定波长为560nm,测定总皂苷含量。

由表2测定结果可知,本发明验药物1组含有丹参酮ⅡA为0.217mg/g,总皂苷含量0.256mg/g,而本发明验药物2组含有丹参酮ⅡA为0.203mg/g,总皂苷含量0.489mg/g。
本发明中药组合物的有益效果:
本发明中药组合物以中医药理论为指导,组方选方合理,诸药相配,使得处方具有行气止痛,化瘀通络,补气养血,化痰开窍之功。本发明中药组合物中有效成分(丹参酮ⅡA及总皂苷含量)明确,其中丹参酮ⅡA现代药理学证明其具有活血化瘀作用,从而为本发明内在质量提高提供实验依据。且本发明生产制备工艺科学简单可行,生产成本较低,具有产业化大生产的优势。经临床实验验证,本发明中药组合物的总有效率可达91.8%。
本发明降血脂方药的所选的药材,来源,性味归经,功能主治如下:
丹参,为唇形科植物丹参Salvia miltiorrhiza Bge.的根及根茎。性味归经,性微寒,味苦。归心经、肝经。功效与作用,祛瘀止痛、活血痛经、清心除烦。用治月经不调、经闭痛经、癥瘕积聚、胸腹刺痛、热痹疼痛、疮疡肿毒、心烦不眠;肝脾肿大、心绞痛。
姜黄,为姜科植物姜黄Curcuma longaL.的根茎。性味归经,性温,味苦、辛。归脾经、肝经。功效与作用,破血行气、通经止痛。用治心腹痞满胀痛、癥瘕、妇女血瘀经闭、产后瘀停腹痛、跌打损伤、痈肿等。
枳壳,为芸香科植物酸橙Citrus aurantium L.接近成熟的果实。性味归经,性温,味苦、辛、酸。归脾经、胃经。功效与作用,理气宽中、行滞消胀。用治胸胁气滞、胀满产痛、食积不化、痰饮内停、胃下垂、子宫脱垂、脱肛等。
鸡血藤,为豆科植物密花豆Spatholobus suberects Dunn.的藤茎。性味归经,性温,味苦、甘。归肝经、肾经。功效与作用,补血、活血、通络。用治月经不调、血虚萎黄、麻木瘫痪、风湿痹痛。
黄芪,豆科植物蒙古黄芪Astragalus membranaceus Bge.var.mongholicus(Bge.)Hsiao和膜荚黄芪A.membranaceus(Fisch.)Bge.的根。性味归经,性微温,味甘。归脾经、肺经。功效与作用,补气固表、利尿、托毒排脓、生肌。治疗气短心悸、乏力、虚脱、自汗、盗汗、体虚浮肿、慢性肾炎、久泻、脱肛、子宫脱垂、痈疽难溃、疮口久不愈合、小儿支气管哮喘、慢性乙型肝炎、慢性肾炎和病毒性心肌炎。补气宜炙用,止汗、利尿、托毒排脓生肌宜生用。
毛冬青,为冬青科植物毛冬青的根。夏、秋采,切片晒干。性味,《广西中草药》味微苦甘,性平,无毒。功能主治,清热解毒,活血通脉。治风热感冒,肺热喘咳,喉头水肿,扁桃体炎,痢疾,冠心病,脑血管意外所致的偏瘫,
天麻,为兰科植物天麻Gastrodia elata Bl.的块茎。性味归经,性平,味甘。归肝经。功效与作用,平肝息风止痉。用治头痛眩晕、肢体麻木、小儿惊风、癫痫抽搐、破伤风。
大黄,蓼科植物掌叶大黄Rheum palmatum,L.、唐古特大黄Rheum tanguticumMaxim.ex Balf.或药用大黄Rheumoj-flcinale Baill.的干燥根和根茎。性味归经,性寒,味苦。归脾经、胃经、大肠经、肝经、心包经。功效与作用,泻热通便、凉血解毒、逐瘀通经。用治实热便秘、积滞腹痛、泻痢不爽、湿热黄疸、血热吐衄、目赤、咽肿、肠痈疔疮、瘀血经闭、跌打损伤。外用适量,治水火烫伤、上消化道出血。
丝瓜络,为葫芦科植物丝瓜Luffa cylindrica(L.)Roem.的成熟果实的维管束。性味归经,性平,味甘。归肺经、胃经、肝经。功效与作用,活血、通络、祛风。用治各种原因引起的气血瘀滞、脉络不通的胸胁疼痛、腹痛、腰痛、睾丸疼痛、妇人经痛、乳汁不通等;肺热咳嗽、痔瘘、崩漏等。

石菖蒲,天南星科植物石菖蒲Acorus tatarionwii Schott.的根茎。性味归经,性微温,味辛、苦。归心经、胃经。功效与作用,开窍、豁痰、理气、活血、散风、去湿。用治癫痫、痰厥、热病神昏、健忘、气闭耳聋、心胸烦闷、胃痛、腹痛、风寒湿痹、痈疽肿毒、跌打损伤等。
泽泻,泽泻科植物泽泻Alisma plantago-aquatica L.var.orientalis(Sam.)Juzep.的块茎。性味归经,性寒,味甘、淡。归肾经、膀胱经。功效与作用,利小便、清湿热。用治小便不利、水肿胀满、呕吐、泻痢、痰饮、脚气、淋病、尿血等。
桑寄生,桑寄生科植物桑寄生Taxillus chinensis(DC.)Danser.的带叶茎枝。性味归经,味苦、甘,性平。归肝经、肾经。功效与作用,补肝肾、强筋骨、祛风湿、安胎元。用治风湿痹痛、腰膝酸软、筋骨无力、崩漏经多、妊娠漏血、胎动不安、高血压。
具体实施方式
以下是本发明内容的具体实施例,用于阐述本申请文件中所要解决技术问题的技术方案,有助于本领域技术人员理解本发明内容,但本发明技术方案的实现并不限于这些实施例。
实施例1
取丹参150g、姜黄150g、枳壳150g、鸡血藤150g、黄芪100g、毛冬青100g、天麻90g、大黄90g、丝瓜络90g、桂枝90g、僵蚕60g、全蝎60g,采用真空微波干燥法进行干燥,干燥温度为65℃,干燥时间为1小时,并采用普通粉碎成100目细粉,混合均匀后,再加入药剂学上常用的淀粉制成胶囊剂,即得。
实施例2
取丹参150g、姜黄150g、枳壳150g、鸡血藤150g、黄芪100g、毛冬青100g、天麻90g、大黄90g、丝瓜络90g、桂枝90g、僵蚕60g、全蝎60g、伸筋草60g、当归60g、络石藤60g、刺五加60g、石菖蒲60g、泽泻60g、桑寄生60g,采用真空微波干燥法进行干燥,干燥温度为65℃,干燥时间为1小时,并采用普通粉碎成100目细粉,混合均匀后,再加入药剂学上常用的淀粉制成胶囊剂,即得。
实施例3
取丹参160g、姜黄140g、枳壳140g、鸡血藤160g、黄芪90g、毛冬青110g、天麻100g、大黄100g、丝瓜络80g、桂枝100g、僵蚕70g、全蝎50g,采用真空微波干燥法进行干燥,干燥温度为70℃,干燥时间为0.5小时,并采用普通粉碎成120目细粉,混合均匀后,再加入药剂学上常用的辅料制成各种不同口服制剂,即得。
实施例4
取丹参140g、姜黄160g、枳壳160g、鸡血藤140g、黄芪110g、毛冬青90g、天麻80g、大黄80g、丝瓜络100g、桂枝80g、僵蚕50g、全蝎70g,采用真空微波干燥法进行干燥,干燥温度为60℃,干燥时间为2小时,并采用普通粉碎成80目细粉,混合均匀后,再加入药剂学上常用的辅料制成各种不同口服制剂,即得。
实施例5
取丹参160g、姜黄140g、枳壳160g、鸡血藤160g、黄芪110g、毛冬青90g、天麻80g、大黄100g、丝瓜络80g、桂枝100g、僵蚕50g、全蝎50g、伸筋草70g、当归70g、络石藤50g、刺五加50g、石菖蒲70g、泽泻70g、桑寄生50g,采用真空微波干燥法进行干燥,干燥温度为65℃,干燥时间为1小时,并采用普通粉碎成100目细粉,混合均匀后,再加入药剂学上常用的辅料制成各种不同口服制剂,即得。
实施例6
取丹参140g、姜黄160g、枳壳140g、鸡血藤140g、黄芪90g、毛冬青110g、天麻100g、大黄80g、丝瓜络100g、桂枝80g、僵蚕70g、全蝎70g、伸筋草50g、当归50g、络石藤70g、刺五加50g、石菖蒲50g、泽泻50g、桑寄生70g,采用真空微波干燥法进行干燥,干燥温度为65℃,干燥时间为1小时,并采用普通粉碎成120目细粉,混合均匀后,再加入药剂学上常用的辅料制成各种不同口服制剂,即得。
为了进一步的验证本发明降血脂制剂的临床疗效,我们将上述具体实施例1-2中制备的药物进行了相应的临床试验,现将结果报告如下。
二、临床观察及疗效
2.1、研究对象:
病例选择:标准根据《中药新药治疗高脂血症的临床指导原则》指定诊断标准即:血清总胆固醇(TC)>5.32mmol/L,甘油三酯(TG)≥1.7mmoL/L,或高密度脂蛋白胆固醇(HDL-C):男<1.04mmol/L,女<1.17mmol/L。全部病例均为饮食控制无效的病人,患者临床多表现为眩晕、肢重、胸闷、头痛、心悸、气促等症,脉象弦滑或弦多见,少有结代脉;舌质以暗红、紫暗多见,舌体胖,舌苔以腻苔多见。中医辨证以血瘀和痰瘀互结型多见。
2.2、研究方法
⑴一般资料:收集符合上述病例标准的高脂血症患者119例,分为两组,治疗1组58例,男31例,女27例;年龄45~69岁,平均54.3±1.4岁;病程2~6年,平均病程4.3±0.5年,治疗2组61例,男33例,女28例;年龄43~66岁,平均52.7±1.6岁;病程2~7年,平均病程4.6±0.7年,两组患者之间年龄、性别,病程无明显差异,具有可比性。
⑵药物组:
本发明药物组:分别按照如上述实施例1组、2组中所述的原料配比及制备方法,制备成胶囊剂,即得。
2.3、治疗方法与疗效
⑴治疗方法:
试验1组给予制备实施例1组的胶囊,试验2组给予制备实施例2组的胶囊,每日3次,分早、中、晚各1次温服,每次3粒/次,连服3个月后;服药前均测定空腹血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)等各项血脂指标,治疗结束后第1天空腹抽血复查空腹血清各项血脂指标。
⑵疗效标准:参照《中药新药治疗高脂血症的临床研究指导原则》,显效:血脂各项指标达到以下任何一项者,TC下降≥20%或TG下降≥40%,或HDL-C上升0.25mmol/L;有效:血脂检查达到以下任何一项者,TC下降10%~20%或下降20%~40%或LDL-C下降10%~20%或HDL-C上升0.1~0.26mmol/L;无效:未达到上述标准。
⑶统计学分析:各组之间对比采用t检验与X2检验。
实验结果:
表3两组患者血脂指标水平的比较
注:治疗前后比较,▲▲P<0.01,▲P<0.05。
从表3结果可以看出,患者服用本发明降血脂药物后,试验1组和2组的患者空腹血清中总胆固醇(TC)、甘油三酯(TG)的数值下降明显,具有统计学意义(P<0.05),但实验1组和2组患者血清中低密度脂蛋白胆固醇(LDL-C)LDL-C高密度脂蛋白胆固醇(HDL-C)含量升高,但无均无统计学差异。其中试验2组的疗效最佳。该实验说明,提示本发明中药组合物对高血症患者有较好的治疗作用。
表4两组临床治疗效果比较
注:试验1组比较,▲▲P<0.01,▲P<0.05。
从表4结果可知,本发明试验1组和2组总有效率分别为63.8%、91.8%,上述临床数据表明,本发明中药组合物在临床应用具有显著的治疗效果,对于高血脂患者,药物在使用过程中,未发现对有任何毒副作用。
综上可知,本发明中药组合物的制备工艺简单,可行,适于工艺化大生产,具有很好的应用前景。
Claims (4)
1.一种降血脂的中药组合物,其特征在于,它包括如下重量配比的原料药组成:丹参140~160份、姜黄140~160份、枳壳140~160份、鸡血藤140~160份、黄芪90~110份、毛冬青90~110份、天麻80~100份、大黄80~100份、丝瓜络80~100份、桂枝80~100份、僵蚕50~70份、全蝎50~70份。
2.如权利要求1所述的中药组合物,其特征在于,它包括如下重量配比的原料药组成优选为:丹参150份、姜黄150份、枳壳150份、鸡血藤150份、黄芪100份、毛冬青100份、天麻90份、大黄90份、丝瓜络90份、桂枝90份、僵蚕60份、全蝎60份。
3.如权利要求1或2所述的中药组合物,所述中药组合物的制备方法步骤如下:取上述药材,采用真空微波干燥法进行干燥,干燥温度为60~70℃,干燥时间为0.5-2小时,并采用普通粉碎成80~120(目)细粉,混合均匀后,再加入药剂学上常用的辅料制成各种不同口服制剂,即得。
4.如权利要求1所述的中药组合物在制备治疗高血脂疾病药物中的应用。
