Abstract
本发明公开了一种用于咳嗽、痰多、气喘的组合物颗粒剂及其制备方法。该组合物颗粒剂包括以下重量份的原料:橘红90‑110份、杏仁70‑90份、桑叶40‑60份、桔梗40‑60份、橘皮40‑60份、茯苓20‑40份、紫苏子20‑40份、甘草10‑30份、赋形剂50‑60份、黏合剂15‑25份、润滑剂10‑12份、防腐剂5‑8份。该组合物颗粒剂是通过提取、浓缩、制粒、整粒、干燥等步骤制备的。该组合物颗粒剂适用于伤风感冒及风寒引起的咳嗽、痰多,气喘,具有吸收快、作用迅速,携带方便等特点,老少皆宜。

技术领域
本发明属于生物医药领域,涉及一种组合物颗粒剂及其制备方法,特别是涉及一种用于咳嗽、痰多、气喘的组合物颗粒剂及其制备方法。
背景技术
咳、痰、喘是下呼吸道感染的常见主征。当呼吸道发生炎症病变时,呼吸道液体的成分就会发生改变,形成痰液。痰液中包含粘液、异物、病原微生物,各种炎症细胞及坏死脱落的粘膜上皮细胞等成分。中医将痰分为寒痰、风痰、热痰、湿痰及燥痰等,形成痰的原因有很多,第一,体质虚弱,中气不足,由于脾虚不运,可以使水湿停留,凝聚为痰;第二,脾胃薄弱,宿滞逗留,损伤脾胃;或恣食生食、瓜果,中阳被伤;或因热病饮水过多,脾运不及等等都可以使水湿停留,聚集为痰。还有恣食肥甘,胃中浊气郁蒸,酿湿生热也可以化为痰浊;第三,外感失治,或体弱屡患外感,肺气被伤,不能输津四布,通调水道下输膀胱,使水液停留,也可成痰,痰贮于肺,肺气不利,痰涌气道而发,必致咳嗽痰多。风寒侵袭机体,轻者束表,证候恶寒重,发热轻,无汗,鼻塞声重或鼻痒喷嚏,时流清涕,咽痒,咳嗽,咳痰稀薄色白,舌淡红舌苔薄白而润,脉浮或浮紧。重者袭肺,可见咳嗽、咽痒、鼻塞、流涕、多嚏,恶风、咳痰色白质稀,舌淡红苔白,脉浮或浮紧等症状。风寒犯肺可见咳嗽痰多粘腻色白,喘而胸满,以痰湿蕴肺为主。
目前,具有止咳化痰、平喘功效的药品种类繁多,剂型以口服制剂为主,包括糖浆剂、片剂、口服液、丸剂等。大多数患者选择糖浆剂,主要是因为其口感适宜,但是糖浆剂中含有大量的糖,在制备和贮藏过程中极易被微生物污染,导致糖浆霉败变质。片剂虽然稳定性较糖浆剂强,但是也存在幼儿及昏迷病人不易吞服、压片时加入的辅料有时影响药物的溶出和生物利用度等缺点。口服液由于存在首关效应会导致生物利用度低,丸剂也会存在服用不便的缺点。风寒引起的痰多患者涉及各个年龄层,因此有必要制备疗效好、服用方便的用于治疗风寒引起的痰多、气喘等呼吸道感染的药物以满足各个年龄层患者的需要。
中药是我国特有的中医药理论与实践的产物,资源丰富,历史悠久,是人类最早应用的纯天然有机物质,其在治疗疾病方面有着毒副作用小、疗效好且作用持久的优势。
橘红为芸香科植物橘Citrus reticulata Blanco及其栽培变种的干燥外层果皮。收载于《中国药典》2015年版:【性味与归经】辛、苦,温。归肺、脾经。【功能与主治】理气宽中,燥湿化痰。用于咳嗽痰多,食积伤酒,呕恶痞闷。
杏仁为蔷薇科植物山杏Armeniaca vulgaris Lam.var.ansu(Maxim.)Yu et Lu.、西伯利亚杏Armeniaca sibirica(L.)Lam.、东北杏Armeniaca mandshurica(Maxim.)Skv.或杏Armeniaca vulgaris Lam.的干燥成熟种子。收载于《中药大辞典》:【性味与归经】苦,温,有毒。入肺、大肠经。【功能与主治】祛痰止咳,平喘,润肠。治外感咳嗽,喘满,喉痹,肠燥便秘。
桑叶为桑科植物桑Morus alba L.的干燥叶。收载于《中国药典》2015年版:【性味与归经】甘、苦,寒。归肺、肝经。【功能与主治】疏散风热,清肺润燥,清肝明目。用于风热感冒,肺热燥咳,头晕头痛,目赤昏花。
桔梗为桔梗科植物桔梗Platycodon、grandiflorum(Jacq.)A.DC.的干燥根。收载于《中国药典》2015年版:【性味与归经】苦、辛,平。归肺、经。【功能与主治】宣肺,利咽,祛痰,排脓。用于咳嗽痰多,胸闷不畅,咽痛音哑,肺痈吐脓。
橘皮为芸香科植物福橘或朱橘等多种橘类的果皮。收载于《中药大辞典》:【性味与归经】辛,苦,温。入脾、肺经。【功能与主治】理气,调中,燥湿,化痰。治胸腹胀满,不思饮食,呕吐哕逆,咳嗽痰多。
茯苓为多孔菌科真菌茯苓Poriacocos(Schw.)Wolf的干燥菌核。收载于《中国药典》2015年版:【性味与归经】甘、淡,平。归心、肺、脾、肾经。【功能与主治】利水渗湿,健脾,宁心。用于水肿尿少,痰饮眩悸,脾虚食少,便溏泄泻,心神不安,惊悸失眠。
紫苏子为唇形科植物紫苏Perilla/rwiescem'(L.)Britt.的干燥成熟果实。收载于《中国药典》2015年版:【性味与归经】辛,温。归肺经。【功能与主治】降气化痰,止咳平喘,润肠通便。用于痰壅气逆,咳嗽气喘,肠燥便秘。
甘草为豆科植物甘草GZwr/ikauralensis Fisch.、胀果甘草Glycyrrhizainflata Bat.或光果甘草Glycyrrhizaglabra L.的干燥根和根茎。收载于《中国药典》2015年版:【性味与归经】甘,平。归心、肺、脾、胃经。【功能与主治】补脾益气,淸热解毒,祛痰止咳,缓急止痛,调和诸药。用于脾胃虚弱,倦怠乏力,心悸气短,咳嗽痰多,脘腹、四肢挛急疼痛,痈肿疮毒,缓解药物毒性、烈性。

发明内容
本发明的目的在于提供一种用于咳嗽、痰多、气喘的组合物颗粒剂及其制备方法,以解决现有技术存在的治疗咳嗽、痰多、气喘的药物生物利用度低、口感不佳、服用携带不便等问题。该组合物颗粒剂制备方法简单易操作、原料成本低,具有吸收快、作用迅速、服用和携带方便等优点,口感佳,老少皆宜。
为解决上述问题,本发明采取以下技术方案:
一种用于咳嗽、痰多、气喘的组合物颗粒剂,包括以下重量份的原料:橘红90-110份、杏仁70-90份、桑叶40-60份、桔梗40-60份、橘皮40-60份、茯苓20-40份、紫苏子20-40份、甘草10-30份、赋形剂50-60份、黏合剂15-25份、润滑剂10-12份、防腐剂5-8份。
以上所述赋形剂为糖粉、乳糖、环糊精中的至少一种;所述黏合剂为淀粉浆、羧甲基纤维素钠、羧丙基甲基纤维素中的至少一种;所述润滑剂为硬脂酸镁或滑石粉;所述防腐剂为苯甲酸、山梨酸、山梨酸钾中的至少一种。
以上所述用于咳嗽、痰多、气喘的组合物颗粒剂,在制备用于咳嗽、痰多、气喘的组合物颗粒剂中的用途。
一种以上所述的用于咳嗽、痰多、气喘的组合物颗粒剂的制备方法,包括以下步骤:
S1:按以下重量份称取原料:橘红90-110份、杏仁70-90份、桑叶40-60份、桔梗40-60份、橘皮40-60份、茯苓20-40份、紫苏子20-40份、甘草10-30份、赋形剂50-60份、黏合剂15-25份、润滑剂10-12份、防腐剂5-8份;
S2:取步骤S1称取的原料装入提取罐中,加水煎煮两次,滤过,上清液浓缩成稠膏;
S3:取步骤S2制得的稠膏加入赋形剂、黏合剂、润滑剂、防腐剂混合,搅拌均匀;
S4:将步骤S3的稠膏置于制粒机中制成均匀颗粒;
S5:将步骤S4制成的颗粒置于烤箱中干燥;
S6:待步骤S5干燥后的颗粒冷却,过筛,即得所述组合物颗粒剂。
进一步地,步骤S2第一次加10-15倍量的水煎煮1.5-2h,第二次加8-10倍量的水煎煮1-1.5h,浓缩至相对密度在65℃为1.03-1.07的稠膏;步骤S5中干燥温度为60-80℃,干燥至颗粒含水量低于2%;步骤S6干燥后的颗粒先过10目筛筛除粗大颗粒,再过80目筛筛除细粉。
与现有技术相比,本发明具备如下有益效果:
1.本发明的组合物颗粒剂能有效缓解咳嗽、痰多、气喘,总有效率达94.3%。
2.本发明既保持了汤剂作用迅速的特点,又克服了汤剂临用时煎煮不便的缺点;且味道可口、体积小,服用、贮藏及运输均较方便。
3.本发明原料均为纯中药饮片,安全易得,无毒副作用,制备方法简单易操作。

下面结合具体实施例对本发明作进一步描述,但不限制本发明的保护范围和应用范围:
一、用于咳嗽、痰多、气喘的组合物颗粒剂的制备
一种用于咳嗽、痰多、气喘的组合物颗粒剂的制备方法,包括以下步骤:
S1:按以下重量份称取原料:橘红90-110份、杏仁70-90份、桑叶40-60份、桔梗40-60份、橘皮40-60份、茯苓20-40份、紫苏子20-40份、甘草10-30份、赋形剂50-60份、黏合剂15-25份、润滑剂10-12份、防腐剂5-8份;
所述赋形剂为糖粉、乳糖、环糊精中的至少一种;
所述黏合剂为淀粉浆、羧甲基纤维素钠、羧丙基甲基纤维素中的至少一种;
所述润滑剂为硬脂酸镁或滑石粉;
所述防腐剂为苯甲酸、山梨酸、山梨酸钾中的至少一种;
S2:取步骤S1称取的原料装入提取罐中,加10-15倍量的水煎煮1.5-2h,过滤,药渣加8-10倍量的水煎煮1-1.5h,过滤,合并滤液,滤液浓缩至相对密度在65℃为1.03-1.07的稠膏;
S3:取步骤S2制得的稠膏加入赋形剂、黏合剂、润滑剂、防腐剂混合,搅拌均匀;

S1:按以下重量份称取原料:橘红95份、杏仁75份、桑叶45份、桔梗45份、橘皮45份、茯苓25份、紫苏子25份、甘草15份、赋形剂52份、黏合剂18份、润滑剂11份、防腐剂6份;
所述赋形剂为乳糖;
所述黏合剂为羧甲基纤维素钠;
所述润滑剂为滑石粉;
所述防腐剂为山梨酸;
S2:取步骤S1称取的原料装入提取罐中,加11倍量的水煎煮2h,过滤,药渣加9倍量的水煎煮1.5h,过滤,合并滤液,滤液浓缩至相对密度在65℃为1.04的稠膏;
S3:取步骤S2制得的稠膏加入赋形剂、黏合剂、润滑剂、防腐剂混合,搅拌均匀;
S4:将步骤S3的稠膏置于制粒机中制成均匀颗粒;
S5:将步骤S4制成的颗粒置于烤箱中干燥,设定温度为65℃,干燥至颗粒含水量低于2%;
S6:待步骤S5干燥后的颗粒冷却,先过10目筛筛除粗大颗粒,再过80目筛筛除细粉,剩下的颗粒即为所述组合物颗粒剂。

所述赋形剂为环糊精;
所述黏合剂为羧丙基甲基纤维素;
所述润滑剂为硬脂酸镁;
所述防腐剂为山梨酸钾;
S2:取步骤S1称取的原料装入提取罐中,加12倍量的水煎煮1.8h,过滤,药渣加10倍量的水煎煮1.2h,过滤,合并滤液,滤液浓缩至相对密度在65℃为1.05的稠膏;
S3:取步骤S2制得的稠膏加入赋形剂、黏合剂、润滑剂、防腐剂混合,搅拌均匀;
S4:将步骤S3的稠膏置于制粒机中制成均匀颗粒;
S5:将步骤S4制成的颗粒置于烤箱中干燥,设定温度为70℃,干燥至颗粒含水量低于2%;

S1:按以下重量份称取原料:橘红105份、杏仁85份、桑叶55份、桔梗55份、橘皮55份、茯苓35份、紫苏子35份、甘草25份、赋形剂58份、黏合剂22份、润滑剂11份、防腐剂7份;
所述赋形剂为糖粉和乳糖,其中糖粉38份、乳粉20份;
所述黏合剂为羧甲基纤维素钠和羧丙基甲基纤维素,其中羧甲基纤维素钠10份,羧丙基甲基纤维素12份;
所述润滑剂为滑石粉;
所述防腐剂为山梨酸和山梨酸钾,其中山梨酸3份,山梨酸钾4份;
S2:取步骤S1称取的原料装入提取罐中,加13倍量的水煎煮2h,过滤,药渣加10倍量的水煎煮1.5h,过滤,合并滤液,滤液浓缩至相对密度在65℃为1.06的稠膏;

S1:按以下重量份称取原料:橘红110份、杏仁90份、桑叶60份、桔梗60份、橘皮60份、茯苓40份、紫苏子40份、甘草30份、赋形剂60份、黏合剂25份、润滑剂12份、防腐剂8份;
所述赋形剂为糖粉、乳糖、环糊精,其中糖粉、乳糖、环糊精各20份;
所述黏合剂为淀粉浆、羧甲基纤维素钠、羧丙基甲基纤维素,其中淀粉浆10份、羧甲基纤维素钠7份、羧丙基甲基纤维素8份;
所述润滑剂为滑石粉;
所述防腐剂为苯甲酸、山梨酸、山梨酸钾,其中苯甲酸2份、山梨酸3份、山梨酸钾3份;
S2:取步骤S1称取的原料装入提取罐中,加15倍量的水煎煮2h,过滤,药渣加10倍量的水煎煮1.5h,过滤,合并滤液,滤液浓缩至相对密度在65℃为1.07的稠膏;
S3:取步骤S2制得的稠膏加入赋形剂、黏合剂、润滑剂、防腐剂混合,搅拌均匀;
S4:将步骤S3的稠膏置于制粒机中制成均匀颗粒;
S5:将步骤S4制成的颗粒置于烤箱中干燥,设定温度为80℃,干燥至颗粒含水量低于2%;
S6:待步骤S5干燥后的颗粒冷却,先过10目筛筛除粗大颗粒,再过80目筛筛除细粉,剩下的颗粒即为所述组合物颗粒剂。
二、溶化性试验
分别取实施例1-5样品10g置于烧杯中,每个实施例的样品分别置于两个烧杯中,每个烧杯5g,再分别加入20倍量温度为23-26℃的水和70-80℃的热水,搅拌5min;待冷却至室温,静置一定的时间,进行目测比较,结果见表1。

试验结果表明:本发明实施例1-5的颗粒剂溶化性好。
三、临床试验
1.病例选择
选择痰多、伴有喉痒、咳嗽、气喘等症状的患者。
2.给药方法
试验组:服用本发明实施例3样品,每天3次,饭后服,2天一疗程。
对照组:服用市售某品牌化痰止咳糖浆,每天3次,饭后服,2天一疗程。
3.疗效判定
进行综合判断分痊愈、显效、好转、无效4级标准判定。
①痊愈:无痰,咳嗽喉痒症状消失,气喘症状消失。
②显效:痰量减少或无痰,咳嗽喉痒症状减轻或消失,气喘症状减轻或消失。
③好转:痰量减少,咳嗽喉痒症状减轻,气喘症状减轻。
④无效:痰量不变或增加,咳嗽喉痒症状不变或加重,气喘症状不变或加重。
4.试验结果
试验组70例、对照组50例患者经过一疗程的治疗,结果详见表2。
试验组:痊愈:31.4%,显效:37.2%,好转:25.7%,无效:5.7%,总有效94.3%。

结果表明:采用本发明实施例3的样品,可用于风寒引起的痰多、伴有喉痒、咳嗽、气喘等的治疗,使用安全、有效,有效率高达94.3%,可明显改善痰多、喉痒、咳嗽、气喘等症状,试验过程无过敏和不良反应。试验组痊愈率及显效率均高于对照组。

赵某,男,36岁。感受风寒后咳嗽有痰、咽痒、鼻塞、咳痰色白质稀,舌淡红苔白,脉浮。服用本发明实施例2产品两天后,上述症状消失,停药7天未见反复。

林某,女,46岁。感受风寒后咳嗽有痰、咳痰色白质稀,伴有喉痒、气喘,舌淡红苔白,脉浮紧。服用本发明实施例4产品两天后,上述症状消失,停药7天未见反复。
典型例子3
李某,男33岁。外感风寒后,鼻塞、多嚏,恶风、咳嗽痰多粘腻色白,喘而胸满。服用本发明实施例5产品两天后,上述症状消失,停药7天未见反复。

5.根据权利要求1所述的用于咳嗽、痰多、气喘的组合物颗粒剂,其特征在于,所述防腐剂为苯甲酸、山梨酸、山梨酸钾中的至少一种。
6.一种根据权利要求1-5任一项所述的用于咳嗽、痰多、气喘的组合物颗粒剂的制备方法,其特征在于,包括以下步骤:
S1:按以下重量份称取原料:橘红90-110份、杏仁70-90份、桑叶40-60份、桔梗40-60份、橘皮40-60份、茯苓20-40份、紫苏子20-40份、甘草10-30份、赋形剂50-60份、黏合剂15-25份、润滑剂10-12份、防腐剂5-8份;
S2:取步骤S1称取的原料装入提取罐中,加水煎煮两次,滤过,上清液浓缩成稠膏;
S3:取步骤S2制得的稠膏加入赋形剂、黏合剂、润滑剂、防腐剂混合,搅拌均匀;
S4:将步骤S3的稠膏置于制粒机中制成均匀颗粒;
S5:将步骤S4制成的颗粒置于烤箱中干燥;
S6:待步骤S5干燥后的颗粒冷却,过筛,即得所述组合物颗粒剂。
7.根据权利要求6所述的用于咳嗽、痰多、气喘的组合物颗粒剂的制备方法,其特征在于,步骤S2第一次加10-15倍量的水煎煮1.5-2h,第二次加8-10倍量的水煎煮1-1.5h,浓缩至相对密度在65℃为1.03-1.07的稠膏。
8.根据权利要求6所述的用于咳嗽、痰多、气喘的组合物颗粒剂的制备方法,其特征在于,步骤S5中干燥温度为60-80℃,干燥至颗粒含水量低于2%。
9.根据权利要求6所述的用于咳嗽、痰多、气喘的组合物颗粒剂的制备方法,其特征在于,步骤S6干燥后的颗粒先过10目筛筛除粗大颗粒,再过80目筛筛除细粉。
10.根据权利要求1所述的用于咳嗽、痰多、气喘的组合物颗粒剂,其特征在于,在制备用于咳嗽、痰多、气喘的组合物颗粒剂中的用途。
