本发明公开了一种过敏性鼻炎外用制剂的配方及制备方法,所述外用制剂的成分主要包括由虫草、鹿茸、人参、薄荷、苍耳子、辛夷花经超临界CO2萃取联合水蒸气蒸馏法制得的虫草精油、鹿茸精油、人参精油、薄荷精油、薄荷醇、苍耳子油、辛夷花油以及薄荷脑组合而成。制剂的制备过程主要包括中药材超临界萃取各油状产物、蒸馏法提取混合挥发油与薄荷的均匀混合和分装。本发明的过敏性鼻炎制剂产品能有效治疗过敏性鼻炎,对急、慢性鼻炎,干燥性鼻炎,鼻塞,流鼻涕,打喷嚏,头痛,嗅觉减退,鼻塞不通,伤风感冒,肚痛屙呕,蚊咬止痒,消化不良,汤火灼伤,牙痛头痛,消暑提神,驱风救急,舟车晕浪,消毒止炎等均有一定的治疗效果。

本发明涉及一种过敏性鼻炎外用制剂的配方及制备方法,属于中药提取领域。
背景技术
过敏性鼻炎(AR)是身体对某些过敏原等敏感性增高而在鼻部出现的异常反应。症状主要表现为突然和反复发作性鼻痒、打喷嚏、流涕和鼻塞。过去的20多年中, 随着工业化水平的提高、人类环境的污染,过敏性鼻炎的病人逐年增加,我国中心城市AR的发病率为8.7%-24.1%。目前,大量临床观察表明西药治疗存在一些问题: ① 抗组胺药对一部分患者,特别是合并血管运动性鼻炎的AR患者,不能完全缓解症状,并且难以避免嗜唾等副作用;②局部用激素-尽管疗效好,但是-长期服用,对身体机理的副作用依然较大。因此,寻找副作用小,治疗效果好的治疗过敏性鼻炎制剂并制定合宜的用药方法是目前主要的研究方向。
外治法是中医的重要特色疗法,其以治标治本、无副作用等优点受到患者特别是老年人患者的青睐。经检索中药治疗过敏性鼻炎的相关专利制剂中发现:专利“CN201510283695.3 治疗过敏性鼻炎的中药油剂及其制备方法”该专利将中药成分通过加水煮、加橄榄油煮等工序制成;专利“CN201310197676.X 一种鼻炎/过敏性鼻炎用液体制剂”其中药成分通过加热醇提,提取1-5次后,将溶剂挥发后加入,同时为增溶剂、增稠剂、防腐剂等各种添加剂,以确保产质量;专利“CN2016108611252 治疗过敏性鼻炎的中药组合物、其制备方法及其中药制剂”通过武火、文火煎煮水提,减压浓缩等工序制成。研究发现,治疗过敏性鼻炎的中药制剂制备方法,多选用中药提取技术,而常用的中药提取溶剂提取,成分复杂,溶剂残留等,挥发油提取水蒸气蒸馏法,提取率低,质量控制不稳定。对于受热易分解的有效成分,中药传统的提取技术已不再适用。对于昂贵的中药材采用普通的提取技术提取率低、成本高,而不同提取方法所提取的成分和含量不完全相同,提取方法联合使用可提高中药有效成分的提取率,但相关研究较少。
产品的每一步生产工序均影响成品最终质量,前处理工序研究往往被忽略,例如清洗、干燥,选择合适的工艺技术才能保证中药材质量和减少有效成分损失,并且为简化生产工序、减少生产成本,对于多种药材的提取多采用混合提取法,由于药材有效成分的提取受到提取条件如溶剂、温度、提取时间等因素影响,采用多种药材混合提取很难筛选出满足多种药材提取的最佳条件,多种药材同时提取不仅造成提取效率低,而且成分复杂,有效成分含量低,造成所制备的制剂疗效不显著。
传统的机械叶轮搅拌设备由于有轴、叶轮等转动部件,易磨损疲劳,故障多,生产效率低,且机械搅拌需要有动密封,密封处油污若泄露则对容器内介质或混合物料造成污染,影响产品质量及安全性,机械混合时形成的漩涡严重影响混合的均匀性。
发明内容
本发明的目的在于解决现有技术的不足,提供了一种有效成分含量高、疗效好、效果稳定、制备工序少,使用方便,质量稳定可长期贮存的一种过敏性鼻炎外用制剂及制备方法。
为了实现上述发明目的,本发明采用现代生物工程技术超临界萃取技术,萃取中药材的有效成分精制而成,其主要成分为虫草精油、鹿茸精油、人参精油、薄荷精油、薄荷醇、薄荷脑、苍耳子油、辛夷花油。该过敏性鼻炎外用制剂的制备方法包括如下步骤:将虫草、鹿茸、人参、薄荷、苍耳子、辛夷花中药材洗净,干燥,粉碎过30目筛后,分别利用超临界CO2萃取装置进行萃取获得虫草精油、鹿茸精油、人参精油、薄荷精油、苍耳子油、辛夷花油;再将3/5薄荷精油通过CO2萃取装置进行提纯制备薄荷醇;将上述超临界CO2萃取后的剩余中药渣混合后装入挥发油提取器中,加蒸馏水蒸馏4h,所得挥发油用无水硫酸钠干燥,得混合挥发油。最后将上述萃取产物、蒸馏挥发油与薄荷脑经混合设备充分混合均匀后制得过敏性鼻炎外用油状液体制剂。

与现有技术相比,本专利所述的一种过敏性鼻炎外用制剂及其制备方法具有以下有益效果,本发明采用超声波清洗中药材洗净度高、用水量少,超声波清洗有助于有效成分的分散,进而提高有效成分的提取效率;采用微波干燥技术,干燥效率高,温度低,减少有效成分损失;采用超临界萃取技术和水蒸气蒸馏法联合对各中药材成分进行提取, 充分提取中药材有效成分,提取率显著提高。超临界萃取技术以CO2流体作为萃取剂,通过正交试验分别筛选出各中药材最佳萃取条件,保证各中药材的提取率、有效成分含量稳定,所制备的过敏性鼻炎制剂质量稳定,疗效显著。并且超临界萃取技术具有萃取操作温度低,提取效率高,无毒,无溶剂残留,无二次污染,技术工艺简单,有利于保护目标物质的生物活性等优点。此外,超临界流体对活性成分的高效萃取保证了植物精油中高含量的活性成分。选用液体射流混合器对各有效成分进行混合,混合均匀性好,效率高,对混合容器中液体变动的适应性强。此外,本发明制备的过敏性鼻炎外用制剂为天然本草油,有效成分含量高,治疗效果好,产品质量稳定可控,将适量本产品湿透在棉棒上均匀涂抹于鼻腔内壁,可有效治疗过敏性鼻炎,对于急、慢性鼻炎,干燥性鼻炎,鼻塞,流鼻涕,打喷嚏,头痛,嗅觉减退,鼻塞不通,伤风感冒,肚痛屙呕,蚊咬止痒,消化不良,汤火灼伤,牙痛头痛,消暑提神,驱风救急,舟车晕浪,消毒止炎等均有治疗效果。
具体实施方式
【实施例1】
一种过敏性鼻炎外用制剂选用超临界萃取技术联合水蒸气蒸馏法,提取中药材的有效成分精制而成。其配方按下列重量份比的主要原料加工而成: 虫草10份、鹿茸10份、人参10份、薄荷25份、薄荷脑3份、苍耳子50份、辛夷花40份。所述过敏性鼻炎外用制剂制备方法包括以下步骤:
(1)按重量份比称取虫草、鹿茸、人参、薄荷、苍耳子、辛夷花等中药材分别在功率50W、频率40kHz的条件下超声清洗10min,温度50℃条件下微波干燥时间1h,然后粉碎过30目筛,得中药材粉末,用于步骤(2)至(8)超临界CO2萃取;
(2)将虫草粉末装入超临界CO2萃取装置,在萃取压力20MPa,温度50℃,CO2流量60L/h的条件下萃取1h,收集萃取物即得虫草精油;
(3) 将鹿茸粉末装入超临界CO2萃取装置,在萃取压力20MPa,温度50℃,夹带剂(70%乙醇)用量1ml/g,CO2流量30L/h的条件下萃取3h,收集萃取物即得鹿茸精油;
(4)将人参粉末装入超临界CO2萃取装置,在萃取压力30MPa,温度55℃, CO2流量60L/h的条件下萃取2h,收集萃取物即得人参精油;
(5)将薄荷粉末装入超临界CO2萃取装置,在萃取压力20MPa,温度40℃, CO2流量30L/h的条件下萃取2.5h,收集萃取物即得薄荷精油;
(6)将步骤(5)制得3/5薄荷精油装入超临界CO2萃取装置,在萃取压力9MPa,温度35℃,CO2流量120L/h的条件下萃取1.5h,收集萃取余物即得薄荷醇;
(7)将苍耳子粉末装入超临界CO2萃取装置,在萃取压力25MPa,温度40℃,CO2流量30L/h的条件下萃取2h,收集萃取物即得苍耳子油;
(8)将辛夷花粉末装入超临界CO2萃取装置,在萃取压力35MPa,温度45℃,CO2流量60L/h的条件下萃取1h,收集萃取物即得辛夷花油;
(9)将上述步骤(2)至(8)超临界CO2萃取剩余中药渣混合后装入挥发油提取器中,加蒸馏水蒸馏4h,所得挥发油用无水硫酸钠干燥,得混合挥发油;
(10)将上述步骤(2)至(9)萃取产物虫草精油、鹿茸精油、人参精油、薄荷精油、薄荷醇、苍耳子油、辛夷花油和混合挥发油加入搅拌机中搅拌,搅拌的同时加入薄荷脑搅拌15分钟至搅拌均匀,搅拌均匀后获得油状液体,最后将油状液体分装即得。
【实施例2】
一种过敏性鼻炎外用制剂选用超临界萃取技术联合水蒸气蒸馏法,提取中药材的有效成分精制而成。其配方按下列重量份比的主要原料加工而成: 虫草13份、鹿茸13份、人参13份、薄荷30份、薄荷脑4份、苍耳子55份、辛夷花45份。所述过敏性鼻炎外用制剂制备方法包括以下步骤:
(1)按重量比称取虫草、鹿茸、人参、薄荷、苍耳子、辛夷花等中药材分别在功率50W、频率40kHz的条件下超声清洗10min,温度50℃条件下微波干燥时间1h,然后粉碎过30目筛,得中药材粉末,用于步骤(2)至(8)超临界CO2萃取;
(2)将虫草粉末装入超临界CO2萃取装置,在萃取压力20MPa,温度55℃,CO2流量60L/h的条件下萃取1.5h,收集萃取物即得虫草精油;
(3) 将鹿茸粉末装入超临界CO2萃取装置,在萃取压力25MPa,温度55℃,夹带剂(70%乙醇)用量1ml/g,CO2流量30L/h的条件下萃取3h,收集萃取物即得鹿茸精油;
(4)将人参粉末装入超临界CO2萃取装置,在萃取压力35MPa,温度55℃, CO2流量60L/h的条件下萃取2.5h,收集萃取物即得人参精油;
(5)将薄荷粉末装入超临界CO2萃取装置,在萃取压力25MPa,温度45℃, CO2流量30L/h的条件下萃取2.5h,收集萃取物即得薄荷精油;
(6)将步骤(5)制得3/5薄荷精油装入超临界CO2萃取装置,在萃取压力9.5MPa,温度40℃,CO2流量120L/h的条件下萃取1.5h,收集萃取余物即得薄荷醇;
(7)将苍耳子粉末装入超临界CO2萃取装置,在萃取压力28MPa,温度40℃,CO2流量30L/h的条件下萃取2.5h,收集萃取物即得苍耳子油;
(8)将辛夷花粉末装入超临界CO2萃取装置,在萃取压力40MPa,温度45℃,CO2流量60L/h的条件下萃取1.5h,收集萃取物即得辛夷花油;
(9)将上述步骤(2)至(8)超临界CO2萃取剩余中药渣混合后装入挥发油提取器中,加蒸馏水蒸馏4h,所得挥发油用无水硫酸钠干燥,得混合挥发油;
(10)将上述步骤(2)至(9)萃取产物虫草精油、鹿茸精油、人参精油、薄荷精油、薄荷醇、苍耳子油、辛夷花油和混合挥发油加入搅拌机中搅拌,搅拌的同时加入薄荷脑搅拌15分钟至搅拌均匀,搅拌均匀后获得油状液体,最后将油状液体分装即得。
【实施例3】
一种过敏性鼻炎外用制剂选用超临界萃取技术联合水蒸气蒸馏法,提取中药材的有效成分精制而成。其配方按下列重量份比的主要原料加工而成: 虫草15份、鹿茸15份、人参15份、薄荷35份、薄荷脑5份、苍耳子60份、辛夷花50份。所述过敏性鼻炎外用制剂制备方法包括以下步骤:
(1)按重量比称取虫草、鹿茸、人参、薄荷、苍耳子、辛夷花等中药材分别在功率50W、频率40kHz的条件下超声清洗10min,温度50℃条件下微波干燥时间1h,然后粉碎过30目筛,得中药材粉末,用于步骤(2)至(8)超临界CO2萃取;
(2)将虫草粉末装入超临界CO2萃取装置,在萃取压力20MPa,温度60℃,CO2流量60L/h的条件下萃取2h,收集萃取物即得虫草精油;
(3) 将鹿茸粉末装入超临界CO2萃取装置,在萃取压力30MPa,温度60℃,夹带剂(70%乙醇)用量1ml/g,CO2流量30L/h的条件下萃取3h,收集萃取物即得鹿茸精油;
(4)将人参粉末装入超临界CO2萃取装置,在萃取压力40MPa,温度55℃, CO2流量60L/h的条件下萃取3h,收集萃取物即得人参精油;
(5)将薄荷粉末装入超临界CO2萃取装置,在萃取压力30MPa,温度50℃, CO2流量30L/h的条件下萃取2.5h,收集萃取物即得薄荷精油;
(6)将步骤(5)制得3/5薄荷精油装入超临界CO2萃取装置,在萃取压力9.5MPa,温度45℃,CO2流量120L/h的条件下萃取1.5h,收集萃取余物即得薄荷醇;
(7)将苍耳子粉末装入超临界CO2萃取装置,在萃取压力30MPa,温度40℃,CO2流量30L/h的条件下萃取3h,收集萃取物即得苍耳子油;
(8)将辛夷花粉末装入超临界CO2萃取装置,在萃取压力45MPa,温度45℃,CO2流量60L/h的条件下萃取2h,收集萃取物即得辛夷花油;
(9)将上述步骤(2)至(8)超临界CO2萃取剩余中药渣混合后装入挥发油提取器中,加蒸馏水蒸馏4h,所得挥发油用无水硫酸钠干燥,得混合挥发油;
(10)将上述步骤(2)至(9)萃取产物虫草精油、鹿茸精油、人参精油、薄荷精油、薄荷醇、苍耳子油、辛夷花油和混合挥发油加入搅拌机中搅拌,搅拌的同时加入薄荷脑搅拌15分钟至搅拌均匀,搅拌均匀后获得油状液体,最后将油状液体分装即得。
临床试验
治疗方法
将适量本产品湿透在棉棒上均匀涂抹于鼻腔内壁,每天擦抹4次-5次,7天为一个疗程,21天后观察疗效;
疗效标准
基本治愈:流清涕消失,喷嚏、鼻痒、头痛好转,检查双下甲充血肿胀明显减轻,粘膜色泽趋于正常,双下甲缩小,鼻腔分泌物消失或仅有极少量浆液;
好转:鼻通气好转,夜间仍感鼻通气欠佳,下鼻甲略缩小,粘膜仍充血,少量分泌物;
无效:自觉症状及检查无变化;
结果:选择过敏性鼻炎患者150例,临床症状流鼻涕、打喷嚏、头痛、嗅觉减退、鼻塞不通、鼻腔黏膜潮红或水肿或黏液多等。经过三个疗程的治疗后,对发明对过敏性患者过敏性鼻炎基本治愈率为81.33%(122/150),好转率为16.67%(25/150),总有效率为98.0%。
结果表明,本发明所述治疗过敏性鼻炎的外用制剂对过敏性鼻炎有较好的治疗作用,且能缓解各种过敏性鼻炎的临床症状。
临床应用实例如下:
杜先生, 54岁,患过敏性鼻炎多年,每天早晨起床必狂嚏阵作,严重时鼻涕如清水,鼻塞,伴有头痛。采用本发明所述过敏性鼻炎外用制剂三疗程后症状消失,未复发。
姜女士,28岁,因季节交替,经常感冒引起过敏性鼻炎反复发作,流清涕,头脑昏蒙,检查鼻腔黏膜充血,轻微水肿。采用本发明所述过敏性鼻炎外用制剂三疗程后症状消失,未复发。
李女士,35岁,季节性过敏性鼻炎,鼻塞夜间加重,流鼻涕,打喷嚏,嗅觉减退。采用本发明所述过敏性鼻炎外用制剂三疗程后症状消失,未复发。
龙先生,25岁,鼻腔易受空气质量或天气变化刺激引发过敏性鼻炎,鼻痒,流清涕,鼻腔黏膜红肿有黏液。采用本发明所述过敏性鼻炎外用制剂三疗程后症状消失,未复发。
王先生,34岁,患过敏性鼻炎多年,稍受凉即喷嚏不断,出现鼻痒,鼻塞,流涕等症状,早晨加剧。采用本发明所述过敏性鼻炎外用制剂三疗程后症状消失,未复发。

Claims (15)
1.一种过敏性鼻炎外用制剂的配方,其特征在于主要成分为虫草精油、鹿茸精油、人参精油、薄荷精油、薄荷醇、薄荷脑、苍耳子油、辛夷花油。
2.一种过敏性鼻炎外用制剂的制备方法,包括如下步骤:将虫草、鹿茸、人参、薄荷、苍耳子、辛夷花中药材洗净,干燥,粉碎过30目筛后,分别利用超临界CO2萃取装置进行萃取获得虫草精油、鹿茸精油、人参精油、薄荷精油、苍耳子油、辛夷花油;将3/5薄荷精油通过CO2萃取装置进行提纯制备薄荷醇;将上述超临界CO2萃取后的剩余中药渣混合后装入挥发油提取器中,加蒸馏水蒸馏4h,所得挥发油用无水硫酸钠干燥,得混合挥发油;
最后将上述萃取产物、蒸馏挥发油与薄荷脑经混合设备充分混合均匀后制得过敏性鼻炎外用油状液体制剂。
3.如权利要求2所述的过敏性鼻炎外用制剂的制备方法,其特征在于,所述制剂配方由下列重量份比的主要原料加工而成: 虫草10-15份、鹿茸10-15份、人参10-15份、薄荷25-40份、薄荷脑3-5份、苍耳子50-60份、辛夷花40-50份。
4.如权利要求2所述的过敏性鼻炎外用制剂的制备方法,其特征在于,所述虫草、鹿茸、人参、薄荷、苍耳子、辛夷花中药材质量要求应符合《中华人民共和国药典》。
5.如权利要求2所述的过敏性鼻炎外用制剂的制备方法,其特征在于,所述薄荷脑质量要求应符合GB 1886.199《食品安全国家标准 食品添加剂 天然薄荷脑》。

10.如权利要求2所述的过敏性鼻炎外用制剂的制备方法,其特征在于,所述人参精油超临界CO2萃取最佳条件为萃取压力30-40MPa,温度55℃,萃取时间2-3h,CO2流量60L/h。
11.如权利要求2所述的过敏性鼻炎外用制剂的制备方法,其特征在于,所述薄荷精油超临界CO2萃取最佳条件为萃取压力20-30MPa,温度40-50℃,萃取时间2.5h,CO2流量30L/h。
12.如权利要求2所述的过敏性鼻炎外用制剂的制备方法,其特征在于,所述薄荷醇是以薄荷精油为原料,超临界CO2萃取最佳条件为萃取压力9.0-9.5MPa,温度35-45℃,萃取时间1.5h,CO2流量120L/h,取萃余物得薄荷醇。
