Abstract

本发明公布了一种产粘液细菌的持续发酵装置,包括上下有开口的吊瓶、上盖、有锥形顶部的下盖和收集瓶。利用瓶颈阻隔作用和固体培养基中凝胶特性将细菌与液体培养基分隔开,利用重力作用为细菌提供持续稳定的营养物质,利用细菌产粘液特性实现菌剂收集自动化,利用下盖制造锥形裸露固体培养基便于菌株接种和菌剂收集,选取玻璃、树脂材质可高压蒸汽灭菌,设计形状组合便于灭菌和无菌操作。本发明可用于产粘液细菌的发酵生产,使菌剂持续发酵生产而培养基保持新鲜,实现产粘液细菌发酵过程的节省、持续和便利。

一种产粘液细菌的持续发酵装置
技术领域
本发明涉及微生物设备领域,尤其是一种产粘液细菌的持续发酵装置。
背景技术
琼脂胶等介质可允许培养基营养成分通过,而阻止细菌细胞通过。现有细菌发酵技术采用细菌和培养基液体混合的方式,由于细菌自身分泌抑制物等因素影响,经过一段对数生长期就进入衰亡期,营养物质和细菌分泌的自身抑制物质混在一起,造成无法避免的剩余营养物质的浪费,细菌生长速度也被抑制。有些细菌产生粘液,例如可与豆科作物共生固氮的根瘤菌,根瘤菌与豆科作物共生作用可提高土壤肥力、减少化肥使用、提高粮食品质、保护环境,因而越来越受重视。放线菌等有些菌落与固体培养基紧密结合,而有些细菌菌落则不与固体培养基结合。现有技术难以将微生物生长与营养分隔开来,造成营养物质的浪费和发酵过程的不持续以及耗费劳动较多。
发明内容
本发明的目的是:为了克服现有技术的上述不足,本发明提供一种产粘液细菌的持续发酵装置,用于产粘液细菌的持续发酵。
本发明解决其技术问题所采用的技术方案是:一种产粘液细菌的持续发酵装置,包括吊瓶(1)、上盖(2)、下盖(3)和收集瓶(4),吊瓶(1)由玻璃制成,上下都有开口,中部圆柱形,上盖(2)与吊瓶(1)上口咬合,旋紧上盖(2)可密封吊瓶(1)上口,下盖(3)顶端为圆锥形,空心,树脂制成,与吊瓶(1)下口咬合,旋紧下盖(3)可密封吊瓶(1)下口,旋紧下盖(3),倒入固体培养基,冷却凝固后,旋松下盖(3),可形成裸露的锥形固体培养基,便于接种细菌,收集瓶(4)为敞口烧杯状,玻璃制成,内径比吊瓶(1)中部圆柱形外径小,可搁置吊瓶(1)并收集滴下的细菌粘液菌剂。
本发明的有益效果是:利用吊瓶(1)瓶颈堵住固体培养基,利用下盖(3)锥形顶部制成锥形裸露固体培养基,便于接种细菌,利用固体培养基凝胶(5)分割细菌和培养基营养物质,利用重力作用持续而缓慢地为细菌提供营养物质,利用细菌产粘液性质收集细菌菌剂,本发明可充分利用培养基营养物质,减少营养物质的浪费,方便收集细菌菌剂,减少劳动强度,实现菌剂持续生产而培养基保持新鲜无污染,实现菌剂生产过程的节省、持续和便利。

具体实施方式
图1中,吊瓶(1)为上下有开口圆柱形玻璃瓶状,上盖(2)为树脂质,与吊瓶(1)上口咬合,下盖(3)为树脂质,与吊瓶(1)下口咬合,下盖(3)顶部为圆锥形,收集瓶(4)为圆柱形玻璃烧杯状,内径比吊瓶(1)外径小,便于搁置吊瓶(1)。
本发明的图示实施例的工作过程为:a.将吊瓶(1)、上盖(2)、下盖(3)、收集瓶(4)用高压蒸汽灭菌,灭菌前将下盖(3)拧紧,上盖(2)拧松;b.用琼脂配制固体培养基从吊瓶(1)上口处倒入吊瓶(1)至淹没吊瓶(1)下方瓶颈处;c.固体培养基冷却凝固后旋松下盖(3),凝胶(5)封住吊瓶(1)下口及颈部,并形成裸露的锥形固体培养基;d.将产粘液细菌接种于裸露的锥形固体培养基表面;e.将吊瓶(1)搁置在收集瓶(4)上,从上口往吊瓶(1)中加入液体培养基并旋紧上盖(2);f.细菌细胞随粘液不断下滴,持续生产,吊瓶(1)中培养基保持新鲜无污染。
