Disclosure of Invention
The invention aims to overcome the defects in the prior art and provide a composition for preventing and treating influenza virus, so as to solve the technical problems of drug resistance caused by overdue administration of western medicines for preventing and treating influenza virus, complex compatibility of Chinese medicinal formulas and high cost.
The invention also aims to provide a traditional Chinese medicine compound oral instant membrane preparation and a preparation method thereof, so as to solve the technical problems of unsafe oral instant membrane and poor drug effect caused by poor membrane forming weight.
In order to achieve the above object, according to one aspect of the present invention, there is provided a composition for controlling influenza virus. The composition for preventing and treating influenza virus comprises effective amounts of Dendranthema indicum extract and menthol; wherein the weight ratio of the Dendranthema indicum extract to the menthol is (0.1-0.5): (1-10).
In another aspect of the invention, a traditional Chinese medicine compound oral instant membrane preparation is provided. The traditional Chinese medicine compound oral instant membrane preparation contains the composition for preventing and treating influenza virus, and also contains glucomannan, a softening agent and water, and the total weight of the traditional Chinese medicine compound oral instant membrane preparation is 100 percent, and the content of the components of the Dendranthema indicum extract, the menthol glucomannan, the softening agent and the water is as follows:

weighing raw materials of the components according to the components contained in the traditional Chinese medicine compound oral instant membrane preparation;
adding the weighed shennong chrysanthemum extract, menthol glucomannan and softening agent into the water component for mixing treatment.
Compared with the prior art, the composition for preventing and treating influenza virus takes the effective amount of the extract of the Dendranthema indicum as the functional component, and after the extract is compatible with the menthol, the extract and the menthol can play a role in synergism of administration, so that the composition for preventing and treating influenza virus in the embodiment of the invention has an obvious anti-influenza virus effect, such as an obvious antiviral effect on H1N 1A.

In one aspect, the embodiments of the present invention provide a composition for preventing and treating influenza virus. In one embodiment, the influenza control composition comprises an effective amount of a. shennong chamomilla extract and menthol.
The Dendranthema indicum was discovered in 1982 and was identified as a new variety of Compositae plants, and the chrysanthemum is derived from a wild chrysanthemum variety, and not only has the pharmacological action of the wild chrysanthemum, but also has unique characteristics, such as full-bodied fragrance of flowers, leaves, stems and roots. Researches show that effective amount of the Dendranthema indicum extract has antiviral effect, and after the Dendranthema indicum extract is compatible with menthol, the Dendranthema indicum extract and the menthol can play a role in synergism of administration, so that the embodiment of the inventionThe composition has significant antiviral effect on influenza virus, such as type A H1N1Has obvious antiviral effect.
In one embodiment, the extract of Dendranthema indicum is obtained by extracting under the following extraction conditions:
adopting dry stems and/or leaves of Dendranthema indicum as a raw material, and performing reflux extraction treatment on the raw material; the temperature of the reflux extraction treatment is 60-80 ℃, the pressure is 0.6-0.8 atmospheric pressure, the reflux extraction is carried out for 1-2 hours, and the solvent of the reflux extraction treatment is water. In a specific embodiment, the temperature of the reflux extraction treatment is 80 ℃, the pressure is 0.8 atm, the reflux extraction is performed for 1 hour, and the solvent of the reflux extraction treatment is water.
In a specific embodiment, the method of the reflux extraction process is as follows:
firstly, cleaning dried stems and leaves of Dendranthema indicum and then drying (if the water content is controlled to be less than or equal to 12%);
crushing the dried stems and leaves and sieving the crushed stems and leaves with a 30-mesh sieve;
thirdly, reflux extraction (the reflux extraction temperature is 60-80 ℃, the pressure is 0.6-0.8 atmospheric pressure, the reflux extraction is 1-2 hours, the preferred reflux extraction treatment temperature is 80 ℃, the pressure is 0.8 atmospheric pressure, the soaking is 30 minutes, and the reflux extraction is 1 hour, wherein, the reflux extraction solvent is water, and the material-liquid ratio is that the volume ratio of the raw material to the water is 1 (10-30), specifically 1: 20);
fourthly, filtering and collecting filtrate (if three layers of gauze are adopted for filtering treatment);
fifthly, drying the recovered solution in vacuum (for example, drying until the crude drug ratio is 1:1 or 1: 1.2).
The control of the effective components and content of the Dendranthema indicum extract is realized by controlling the extraction process conditions of the Dendranthema indicum raw material, so that the composition for preventing and treating influenza virus provided by the embodiment of the invention has good clinical effects on influenza virus resistance such as H1N 1A resistance.
In order to effectively extract the anti-influenza virus component contained in the Dendranthema indicum, in a further embodiment, the ratio of the raw material to water is controlled to be 1: 20.

One skilled in the art will appreciate that the actual amount or dose administered and the time course of administration will depend on the nature and severity of the disease being treated, the age and general condition of the subject being treated, and the mode of administration, among other factors.
The menthol has the effects of refreshing, diminishing inflammation and relieving pain of penetrant, can play a synergistic effect on the functional component of the Dendranthema indicum extract, and improves the clinical effect of the Dendranthema indicum extract on resisting influenza viruses. At the same time, it also acts as a flavouring agent due to the specific taste of menthol.
In one embodiment, the weight ratio of the Dendranthema indicum extract to menthol is controlled to be (0.1-0.5): (1-10). The weight ratio of the two is controlled within the range, so that the two play a synergistic effect, and the clinical effect of the Dendranthema indicum extract on resisting influenza viruses is improved.
Therefore, the influenza virus prevention and treatment composition provided by the embodiment of the invention takes an effective amount of the dendranthema indicum extract as a functional component, and the effective amount of the dendranthema indicum extract and the menthol are compatible, so that the dendranthema indicum extract and the menthol can play a synergistic effect of administration, and therefore, the influenza virus prevention and treatment composition provided by the embodiment of the invention has an obvious anti-influenza virus effect, is safe to use, and does not generate drug resistance.
On the basis of the above embodiments, the influenza virus prevention and treatment composition according to the embodiments of the present invention may further include a pharmaceutically acceptable adjuvant, which refers to any adjuvant known to those skilled in the art to be suitable for a specific administration mode. As in particular embodiments, the pharmaceutically acceptable excipients may include one or more pharmaceutically acceptable carriers, solvents, excipients, buffers, stabilizers, adjuvants, prodrugs and other therapeutic agents whose treatment benefits from or enhances the above-described influenza virus control compositions that are compatible with the functional components shennong chrysanthemum extract and menthol. It is to be understood that each of the adjuvants described above should be non-toxic, not interfere with or impair the function and action of the influenza virus control compositions of the embodiments of the present invention. In addition, the auxiliary materials can be selected flexibly according to the existing preparation formulation of the influenza virus prevention and treatment composition in the embodiment. In one embodiment, the excipient is a film-forming agent.
Therefore, on the other hand, the embodiment of the invention also provides a traditional Chinese medicine compound oral instant membrane preparation. In one embodiment, the traditional Chinese medicine compound oral instant membrane preparation contains the composition for preventing and treating influenza virus, and also contains components such as glucomannan, a softening agent, water and the like. The traditional Chinese medicine compound oral instant membrane preparation comprises the following components in percentage by weight, based on 100% of the total weight of the traditional Chinese medicine compound oral instant membrane preparation:
the glucomannan contained in the traditional Chinese medicine compound oral instant membrane preparation has the characteristics of water solubility, gel solubility, membrane forming property, rheological property and the like, so that the glucomannan is used as a membrane forming agent in the embodiment of the invention. The softening agent can assist glucomannan to form a film, improve the film forming quality of the traditional Chinese medicine compound oral instant film preparation, endow the traditional Chinese medicine compound oral instant film preparation with good tape casting film forming performance, ensure that the formed film is flat, smooth, soft and tough, safe and nontoxic, and ensure that the composition for preventing and treating influenza virus is compatible with the film forming agent, so that the composition for preventing and treating influenza virus such as the Dendranthema indicum extract and menthol has good release degree.
In one embodiment, the softening agent is at least one of lecithin, glycerin and sorbitol. By selecting the softening agent, the synergistic effect of film forming can be exerted between the softening agent and glucomannan, the film forming quality is improved, the compatibility of the composition for preventing and treating influenza virus, such as the Dendranthema indicum extract and the menthol, with the film forming agent is improved, and the release degree of the composition for preventing and treating influenza virus is improved.
The water component contained in the Chinese medicinal compound oral instant membrane preparation should be water meeting the medical field standard, such as purified water, distilled water, etc.
Therefore, the compound oral instant membrane preparation of the embodiment of the invention adopts the composition for preventing and treating influenza virus as an effective functional component, and has an obvious anti-influenza virus effect. The composition has good casting film forming property, is soft and tough, is safe and nontoxic, and can ensure that the composition for preventing and treating influenza virus has good release degree and high compatibility with a film forming agent.
In another aspect, on the basis of the compound oral instant membrane preparation in the embodiments of the present invention, the embodiments of the present invention provide a preparation method of the compound oral instant membrane preparation. In one embodiment, the preparation method of the compound oral instant membrane preparation comprises the following steps:
step S01: weighing raw materials of the components according to the components contained in the traditional Chinese medicine compound oral instant membrane preparation;
step S02: adding the Dendranthema indicum extract, the menthol glucomannan and the softening agent which are weighed in the step S01 into the water component for mixing treatment.
The components and the contents of the components of the compound oral instant film preparation of traditional Chinese medicine described in the above step S01 are as described in the above compound oral instant film preparation of traditional Chinese medicine, and are not described herein again for the sake of brevity.
The mixing treatment in step S02 may be a conventional mixing method of medicinal materials, or may be a stirring and mixing method directly, or may be an ultrasonic treatment method, and any method may be used as long as it is a sol that can achieve uniform dissolution or dispersion of each component in water to form a stable dispersion system.
In a preferred embodiment, the method for mixing the weighed shennong chrysanthemum extract, menthol glucomannan and softening agent into the water component in the step S02 is as follows:
dissolving the glucomannan in water, adding water while stirring until the glucomannan is completely dissolved into a sol state, and obtaining glucomannan sol;
dissolving the menthol in water to prepare menthol aqueous solution;
heating 1% -2% sodium hydroxide solution to 85-100 deg.C, adding softener under stirring, cooling to 30-40 deg.C, filtering, and controlling pH of filtrate to 8-9 to obtain softener emulsion; in one embodiment, the softening agent is at least one of lecithin, glycerin and sorbitol.
Dissolving the Dendranthema indicum extract with water to prepare an aqueous solution of the Dendranthema indicum extract;
adding the menthol aqueous solution into glucomannan sol for mixing treatment, and then adding the lecithin emulsion for continuously mixing treatment.
Wherein, the steps of obtaining glucomannan sol, preparing menthol aqueous solution, obtaining lecithin emulsion and preparing shennong chrysanthemum extract aqueous solution have no requirement of sequence. Through the mixing treatment mode, the components can be uniformly dispersed to form stable sol, so that the film forming quality of the traditional Chinese medicine compound oral instant film is improved, the stable release of the anti-influenza virus and influenza virus prevention and control composition is improved, and the drug effect stability of the traditional Chinese medicine compound oral instant film preparation is improved.
Therefore, the preparation method of the traditional Chinese medicine compound oral instant membrane in the embodiment of the invention ensures that the traditional Chinese medicine compound oral instant membrane preparation has stable efficacy, safety, no toxicity, low cost and good industrial feasibility.
The invention is further explained by specific embodiments of the composition for preventing and treating influenza virus, the traditional Chinese medicine compound oral instant membrane and the preparation method thereof.
Example of composition for controlling influenza virus
