Abstract
本发明涉及将作为抗菌剂的椰油醇聚醚硫酸酯锌,其用于治疗与疮疱丙酸杆菌相关的皮肤病症。
Description
技术领域
本发明的领域涉及椰油醇聚醚硫酸酯锌对抗细菌痤疮丙酸杆菌的新的抗菌性能;及其在用于与痤疮丙酸杆菌相关的痤疮和皮肤失调的治疗的化妆品领域以及皮肤病学领域的应用。
背景技术
皮脂是皮肤皮脂腺的分泌物油脂膜,用于保护皮肤,并与汗水混合避免皮肤干燥。
皮脂使得皮肤防水,参与表皮结构的发育。其避免微生物对其酸化(在乳酸和脂肪酸的存在下),并保证一定程度的不渗透性。其使得皮肤柔软并参与表皮的发育。
皮脂通常通过毛囊孔到达皮肤表面。
在痤疮期间,毛囊漏斗部中的过量皮脂呈现有利于痤疮丙酸杆菌定植的环境。痤疮病变的外观取决于由于过高密度的痤疮丙酸杆菌,通过先天免疫受体的过强烈的促炎症反应。
这些细菌事实上具有通过释放引起组织炎症的脂肪酸,而代谢皮脂的甘油三酯的特质。
出乎意料且令人惊讶地,本申请人证实了椰油醇聚醚硫酸酯锌可以用作对抗痤疮丙酸杆菌的抗菌剂。
发明内容
椰油醇聚醚硫酸酯锌(Zchimmer & Schwarz公司的Zetesol)是一种阴离子表面活性剂,由于固定于金属上的第二条十二烷基链的缘故,其耐受性要比月桂基醚硫酸盐的常规的钠盐高得多。锌分子的存在赋予其对油性皮肤的致病菌非常令人关注的活性,并使其能够充分地限制防腐剂(2%至7%的浓度)。
椰油醇聚醚硫酸酯锌是结合硫酸盐烷基酯和锌(椰子核的亲脂性双链/锌)的新一代表面活性剂。
烷基醚硫酸盐是化妆品和洗浴产品中使用最广泛的表面活性剂。与二元或三元混合物的组合,它们成为明显的起泡剂(不受硬水影响),并具有优异的清洁能力。
根据用于中和它们的阳离子,烷基醚硫酸盐提供了能进一步改善皮肤和眼睛良好的耐受性。
这项对于椰油醇聚醚硫酸酯锌开展的研究能够证明其不仅开发了良好的表面活性剂性能兼具出色的局部耐受性,还提供了其他令人关注的性能。
如烷基醚硫酸盐一样,椰油醇聚醚硫酸酯锌证实了其良好的清洁性能:令人满意的发泡能力,获得柔软且奶油状泡沫,令人愉悦地使用。
然而,因为其潜在的皮肤侵略性比烷基醚硫酸盐少得多而最有可能与其亲脂性双链相关,其与皮肤的相容性表现得令人惊讶。
而且,显示出了水的硬度对于其清洁能力并不重要。
椰油醇聚醚硫酸酯锌具有4.5的酸性pH值,接近于约5.5的皮肤pH值。
因此,其不会使皮肤粗糙,酸性是成人皮肤以及幼儿皮肤的主要特征之一。
椰油醇聚醚硫酸酯锌显示没有任何刺激性和致敏潜力。
椰油醇聚醚硫酸酯锌还证明了其对粘膜良好的耐受性。
这是通过使用鸡胚绒毛膜尿囊膜试验证实的,鸡胚绒毛膜尿囊膜试验的原理是基于,将产品放置于孵化第十天的含胚鸡卵的绒毛膜尿囊膜后,观察产品在五分钟内发生的刺激效果。以1%的椰油醇聚醚溶液进行测试。
在最终pH值5至6,10%和20%浓度的椰油醇聚醚硫酸酯锌水溶液进行细菌学研究。
细菌学研究是对引发易发痤疮皮肤的细菌,痤疮丙酸杆菌进行的:一项在微生物实验室进行的研究证实了这种杀菌功效。这项研究已在成品上证实。
因此,本发明涉及皮肤卫生组合物,其包含与月桂酰羟甲基乙磺酸钠组合的、作为对抗痤疮丙酸杆菌的抗菌剂的椰油醇聚醚硫酸酯锌。根据本发明另一特征,所述组合物作为对抗痤疮丙酸杆菌的抗菌剂是有效的。更具体地,所述组合物旨在用于痤疮的治疗。
本发明的另一方面涉及包含作为对抗痤疮丙酸杆菌抗菌剂的椰油醇聚醚硫酸酯锌组合物的非治疗用途。
不同于某些类似类型的皮肤清洁组合物(其主张必须有盐或者附加的甘草酸或甘草次酸衍生物的存在),本申请目的组合物明确地完全没有这样的盐或附加的衍生物。
优选地(本发明皮肤卫生组合物)在用于易发痤疮皮肤和/或具有痤疮倾向的油性皮肤的治疗和/或卫生化妆品中的用途。
对易发痤疮皮肤和/或具有痤疮倾向的油性皮肤的卫生需要使用适合于组织脆弱性并伴随病理的微生物菌群的产品。
优选地,用于易发痤疮皮肤和/或具有痤疮倾向的油性皮肤清洁产品的开发的框架内,需要满足几个标准:
-癸二脂肪化学成分良好的油脂去除能力
-良好的表面活性剂的耐受性,以不通过回弹效应刺激皮脂腺的方式
-调节皮脂腺功能
-限制相关细菌的生长。
本发明的另一目的涉及一种组合物,更优选地涉及用于易发痤疮皮肤和/或具有痤疮倾向的清洁组合物,其包含作为对抗痤疮丙酸杆菌的抗菌剂的椰油醇聚醚硫酸酯锌,与之结合的
-阴离子表面活性剂月桂酰羟甲基乙磺酸钠;
-和/或选自桃金娘提取物,更具体地,如EP1112079中所述的桃金娘提取物、聚甘油-3单辛酸酯、癸酸甘油酯、西曲溴铵的另一种抗菌剂;
-和/或选自如WO2011/073370中所述的甘油月桂酸酯、蒲葵提取物、南瓜子油、异株荨麻提取物等的抗皮脂溢剂等等。
在本发明的一个特定的实施方案中,根据本发明组合物的赋形基质与阴离子表面活性剂结合实质上作为二元混合物,其中椰油醇聚醚硫酸酯锌可以与月桂酰羟甲基乙磺酸钠组合;且非离子表面活性剂与活性成分以80/20的比例表示。低浓度两性物质的存在可以稳定最终的粘度。
月桂酰羟甲基乙磺酸钠(,Innospec)是具有更好耐受性的阴离子表面活性剂类之一。其在皮肤上使用多年,证实其在儿科领域以及特异反应性领域中,与皮肤优异的生物相容性(5%至10%的浓度)。
发明另一个优选的实施方案中,作为对抗疮疱丙酸杆菌的抗菌剂的椰油醇聚醚硫酸酯锌将与另一种活性成分组合使用:
-如桃金娘提取物,优选地,EP1112079中所述的桃金娘提取物、聚甘油-3单辛酸酯、癸酸甘油酯、西曲溴铵的抗菌剂;
-或如甘油月桂酸酯、蒲葵提取物、南瓜子油、异株荨麻提取物,对痤疮病理有互补作用。
优选地,将在皮肤清洁和消毒组合物中组合阴离子表面活性剂椰油醇聚醚硫酸酯锌和月桂酰羟甲基乙磺酸钠(如)。
进一步地,已知两极性非离子表面活性剂由于它们的脂肪链与氧乙烯链结合成醚形式的分子结构,提供极端柔顺的产品:
-结合环化山梨醇与乙氧基月桂链的聚山梨酯20
-乙氧基氢化蓖麻油(40EO)
-鲸蜡硬脂醇聚醚-60肉豆蔻基甘醇,其脂肪链提供皮肤表面保护。
以EDTA 2Na和柠檬酸在pH=5缓冲,该分子式非常适合油性皮肤pH值。酸性pH与抗炎剂椰油醇聚醚硫酸酯锌联合参与甘油月桂酸酯的抗菌的和抗5-α-还原酶的活性。
优选地,本发明的组合物能局部使用。
更优选地,本发明的组合物具有液体清洁凝胶、起泡剂或气溶胶的形式。
本发明的再一目的为提供一种净化(和/或清洁)易发痤疮皮肤和/或具有痤疮倾向的油性皮肤的美容方法,其特征在于将包含椰油醇聚醚硫酸酯锌的组合物应用于所述的皮肤。
利用以下非限制性实施例,以及构成了本发明化妆品和/或皮肤病学组合物的具体实施方案可以更好的理解本发明。

根据本发明组合物的实施例
实施例1:
月桂酰羟甲基乙磺酸钠(,Innospec)5至20%
椰油醇聚醚硫酸酯锌(Zetesol,Zchimmer & Schwarz)5至20%
椰油酰两性基乙酸二钠3至10%
聚山梨酯200.5至4%
鲸蜡硬脂醇聚醚-60肉豆蔻基甘醇0.5至1.5%
乙氧基氢化蓖麻油0.5%至2%
甘油月桂酸酯0.1至1%
葡萄糖酸锌0.1至0.5%
柠檬酸/EDTA 2Na qsp pH=5
ZETESOL对抗痤疮丙酸杆菌的杀菌活性的评价
评价了随着时间,致病菌数目相对于致病菌初始数目减少的log10(参数logR)。
测试菌株是痤疮丙酸杆菌ATCC6919。
以20%浓度的Zetesol进行测试(即,5%的活性成分椰油醇聚醚硫酸酯锌)。
方法
-设置以使待测试产品与以滴定测试的约106CFU/ml的痤疮丙酸杆菌悬浮液接触1、5、10和15分钟。
-各个接触持续时间后,对1ml的培养琼脂样品的内含物进行活菌计数(哥伦比亚)。
-在厌氧条件下36℃孵育72小时。
-通过直接计数进行剩余细菌的活菌计数。
以log10计算下降。
对平皿进行计数,并且将这个结果与CFU/ml的数值进行比较。
CFU/ml下降数值的log10的计算根据下式进行:
对数减少=Log10(现有CFU的数值TO/剩余CFU的数值Tx)
表1所示的结果是两次独立实验的平均值(除了持续时间10分钟的,其只进行一次试验)。另请参见图1。

良好的杀菌活性从接触的第一分钟就恰当地观察到了,所述时间接近于用作冲洗产品的使用时间。
该活性在15分钟最佳(达到的最大可视阈值)。
实施例2:
根据本发明的另一组合物实例实施根据上文提到的相同的方案的杀菌活性同样的评价。
所述测试的组合物满足下列配方:
-Zetesol Zn(浓度20%,即活性成分椰油醇聚醚硫酸酯锌5%)
-Iselux(浓度10%,即活性成分月桂酰甲基羟乙基磺酸钠8%)
-鲸蜡硬脂醇聚醚-60肉豆蔻基甘醇,增稠剂
-椰油酰两性基乙酸Na-两性表面活性剂
-辛乙二醇
-乳酸
-氢氧化钠
-适量水
获得的结果示于下文表2中:

在1分钟时观察到痤疮丙酸杆菌ATCC6919的菌株1.9log的减少。其显示了的月桂酰羟甲基乙磺酸钠的添加能够显著改善椰油醇聚醚硫酸酯锌对抗痤疮丙酸杆菌的杀菌活性。
对抗痤疮丙酸杆菌的根据实施例1配制产物的杀菌活性的评价
使用和上文相同的方案。
测试的产品:
-根据实施例1的组合物在无菌蒸馏水中稀释至20%=使用冲洗产品稀释。
测试的菌株为
-痤疮丙酸杆菌ATCC6919。
-痤疮丙酸杆菌Sauvage(红霉素耐药菌株R)
-痤疮丙酸杆菌Sauvage(红霉素耐药菌株S)
下文表3所示的结果是两次独立试验的平均值。另请参见图2。

2.根据权利要求1所述的用于皮肤的卫生组合物,其特征在于其进一步包含选自桃金娘提取物、聚甘油-3单辛酸酯、癸酸甘油酯、西曲溴铵的抗菌剂。
3.根据权利要求1或2所述的用于皮肤的卫生组合物,其特征在于其包含选自甘油月桂酸酯、蒲葵提取物、南瓜子油、异株荨麻提取物的抗皮脂溢剂。
