Abstract

COMPOSIÇÕES E MÉTODOS DE RNA CIRCULAR. São descritos no presente documento RNA circular, junto com composições e métodos relacionados. Em algumas modalidades, o RNA circular inventivo compreende fragmentos de íntron de grupo I, espaçadores, um IRES, regiões formadoras de duplex e uma sequência de expressão. Em algumas modalidades, a sequência de expressão codifica um antígeno. Em algumas modalidades, o RNA circular da invenção tem expressão, estabilidade funcional, imunogenicidade, facilidade de fabricação e/ou meia-vida melhoradas quando comparado com o RNA linear. Em algumas modalidades, os métodos e construtos inventivos resultam em eficiência de circularização, eficiência de splicing e/ou pureza melhoradas quando comparados com abordagens de circularização de RNA existentes.
