Abstract
La presente invención se refiere a un método de tecnología novedosa que ofrece eficiencia no paralela, flexibilidad, utilidad y velocidad para el descubrimiento y optimización de polipéptidos que tienen especificidad y/o funcionalidad de unión deseada, que incluye moléculas de unión al antígeno tales como anticuerpos y fragmentos de los mismos, para los fenotipos funcionales y/o de unión deseados. El método novedoso se base en constructos de transposición y diversas bibliotecas de ADN clonadas en vectores de transposición y su transfección en células huésped por expresión temporal concomitante de una enzima de transposasa funcional. Esto asegura una introducción eficiente y estable de los vectores de expresión basados en transposón en células huésped de vertebrado en una etapa, que pueden clasificarse entonces por un fenotipo funcional o de unión deseado de las proteínas expresadas, después de lo cual las secuencias de codificación relevantes para las proteínas expresadas, que incluyen anticuerpos y fragmentos de los mismos, pueden identificarse por técnicas de clonación y secuenciación de ADN estándar.
