Abstract:
Remarkable attention has been attracted to Chondroitinase ABCI (cABCI) enzyme due to its application in the treatment of spinal cord injuries. Low thermal stability limits the utilization of this enzyme as a therapeutic way. The immobilization of cABCI onto the Fe3 O4@dextran nanoparticles was used in this study to enhance its thermal stability. In this research, the factors influencing the activity of cABCIbound to the dextran-coated Fe3O4 nanoparticles were assessed by the response surface methodology, and the optimal conditions were obtained as follows: pH 6.3, temperature 24 ˚C, enzyme/support mass ratio 1.27, and incubation time 5.7 h. The results indicated that attachment of cABCI tothe Fe3 O4@dextran support can significantly improve the thermal stability. The in-vitro study of this enzyme also revealed that 70% of the cABCI loaded on the support was released after 9 h.
