Abstract
Description
1.起源および寄託
本株は、英国Bradfordの土壌から単離され、2003年3月5日にNational Collection of Industrial and Marine Bacteria(NCIMB)に寄託され、ブダペスト条約の下に登録番号第NCIMB41164号が指定された。
2.形態学的および培養的特徴性
(1)多型増殖
(2)運動性:不動性
(3)非胞子形成種
(4)グラム陽性
(5)好気性
(6)栄養寒天上での増殖は、30℃で48時間以内にサーモンピンク色の円形コロニーを与える。
(1)シュードモナス・フロレセンス(P. florescens)、サッカロマイセス・セレビシエ(S. cerevisiae)およびアスペルギルス・テレウス(A. tereus)を、30℃の栄養ブロス中で、後期指数増殖期に到達するまで培養した。得られた培養ブロスを遠心分離して、バイオマスを除去し、上清を残した。
(2)25%アクリルアミド溶液を調製した。この溶液に、10ppmの次亜リン酸ナトリウムおよび1,000ppmの過酸化水素tert−ブチルの溶液を加えた。酢酸を用いて、pHを4.0に調整した。溶液を脱気し、1,000ppmの硫酸アンモニウム第一鉄を加え、その後、重合体を形成させた。
(3)(2)の手順を、(1)に記載した生物の培養体からの上清を用いて繰り返して、水ではなくアクリルアミド溶液を形成させた。得られた溶液重合体の分子量を測定し、水を用いてアクリルアミド溶液を形成するよう調製しておいた重合体と比較した。結果を表1に示す。重合体の分子量は、すべて、実質的に同じであった。
(1)ロドコッカス・ロドクラウスNCIMB41164株を、下記の成分(g/l):リン酸水素二カリウム0.7;リン酸水素カリウム0.3;グルコース1.0;尿素5.0;酵母エキス3.0;硫酸マグネシウム七水和物0.5;塩化コバルト六水和物0.01を含有する培地180lを有する280l入り発酵槽内で増殖させた。培地のpHを、pH7.2に調整した。培養体を、30℃で3日間成長させた。グルコースも、定期的に培養体に仕込んだ。
(2)(1)からの発酵ブロス15lを、プロセス水35lと混合し、次いでこの懸濁液を、水250kgを有する600l入り反応器に充填した。アクリロニトリルを、46.8%のアクリルアミドの濃度が達成されるまで、数時間にわたって反応器に仕込んだ。アクリルアミド溶液25kgを遠心分離して、生体触媒を除去した。アクリルアミド25kgは、遠心分離して生体触媒を除去しなかった。
(3)(2)からの遠心分離したアクリルアミドのサンプル、および遠心分離しなかったアクリルアミドのサンプルを、酸化還元および熱開始剤を用いて、単独重合体として重合させて、約17dl/gのIVを有するゲル重合体を得た。cPでの粘度も測定し、遠心分離および非遠心分離アクリルアミドの双方を用いて製造したサンプルには差がなかった。この重合体の粘度測定の結果(cP)を表2に示す。
20重量%以下の微生物ロドコッカス・ロドクラウスNCIMB41164株の発酵ブロスを含有する30重量%のアクリルアミド溶液を調製した。発酵ブロスを加えたアクリルアミドを、酸化還元および熱開始剤を用いて単独重合体として重合させて、約17dl/gのIVを有するゲル重合体を形成させた。重合体溶液のそれぞれについての一点固有粘度による粘度測定の結果を表4に示す。粘度の結果は、すべて、この重合体に対する仕様セットの範囲内であった。
Claims (18)
- エチレン性不飽和単量体の重合体を製造する方法であって、該単量体が生体触媒反応または発酵工程から得られ、かつ該単量体が細胞性材料および/または発酵ブロスの成分を含み、該エチレン性不飽和単量体または該エチレン性不飽和単量体を含有する単量体混合物を重合させることによって該重合体を形成させ、ここで該細胞性材料および/または該発酵ブロスの成分を該エチレン性不飽和単量体から実質的に除去しない方法。
- エチレン性不飽和単量体に転換することができる基質を供給すること、該基質を生体触媒(この生体触媒は微生物または細胞性材料を含む)に接触させ、それによって該基質を、細胞性材料および場合により発酵の成分を含有するエチレン性不飽和単量体に転換することによって、エチレン性不飽和単量体を製造し、そしてこの工程を細胞の内部または外部で実施し、それを細胞の内部で実施する場合は、場合によりそれが微生物の代謝経路の一部を形成する、請求項1記載の方法。
- 生体触媒が微生物を含み、工程が細胞内部で実施されかつ微生物の代謝過程の一部を形成する、請求項2記載の方法。
- 細胞性材料が細胞全体を含む、請求項1〜3のいずれか一項に記載の方法。
- 細胞性材料が、破砕された細胞性材料を含む、請求項1〜4のいずれか一項に記載の方法。
- 破砕された細胞性材料が、細胞壁材料、細胞膜材料、細胞核材料、細胞質およびタンパク質からなる群から選ばれる、請求項5記載の方法。
- 発酵ブロスの成分が、糖類、多糖類、タンパク質、ペプチド、アミノ酸、窒素源、無機塩類(金属塩を包含する)、ビタミン、成長調節物質、酵素誘導物質、ならびに複合発酵培地成分、例えば、トウモロコシ浸出液および酵母エキスからなる群から選ばれる、請求項1〜6記載の方法。
- エチレン性不飽和単量体が(メタ)アクリルアミドの単量体である、請求項1〜7のいずれか一項に記載の方法。
- 基質が(メタ)アクリロニトリルである、請求項2〜8のいずれか一項に記載の方法。
- 生体触媒がニトリルヒドラターゼ酵素を含む、請求項2〜9のいずれか一項に記載の方法。
- 重合体が(メタ)アクリルアミドの単独重合体または共重合体である、請求項1〜10のいずれか一項に記載の方法。
- エチレン性不飽和単量体が、イタコン酸(またはその塩)、マレイン酸(またはその塩)および(メタ)アクリル酸(またはその塩もしくは誘導体)からなる群から選ばれる、請求項1〜7のいずれか一項に記載の方法。
- 基質が、乳酸、3−ヒドロキシプロピオン酸およびグリセリンからなる群から選ばれる、請求項2〜7または12のいずれか一項に記載の方法。
- 基質を、容器に導入して生体触媒に接触させ、かつ該基質をエチレン性不飽和単量体へと転換して、場合によりその他の単量体を該容器に導入して、単量体混合物を形成させ、該エチレン性不飽和単量体または単量体混合物を、場合により開始剤を該容器に導入することによって、重合条件に付し、それによって該容器内に重合体を形成させる、請求項2〜13のいずれか一項に記載の方法。
- 生体触媒を容器内で生成させる、請求項14記載の方法。
- 生体触媒が、ロドコッカス属の微生物、好ましくはロドコッカス・ロドクラウス種を含む、請求項2〜15のいずれか一項に記載の方法。
- 微生物がロドコッカス・ロドクラウスNCIMB41164である、請求項16記載の方法。
